Designing Calcium Release Channel Inhibitors with Enhanced Electron Donor Properties: Stabilizing the Closed State of Ryanodine Receptor Type 1

New drugs with enhanced electron donor properties that target the ryanodine receptor from skeletal muscle sarcoplasmic reticulum (RyR1) are shown to be potent inhibitors of single-channel activity. In this article, we synthesize derivatives of the channel activator 4-chloro-3-methyl phenol (4-CmC) and the 1,4-benzothiazepine channel inhibitor 4-[-3{1-(4-benzyl) piperidinyl}propionyl]-7-methoxy-2,3,4,5-tetrahydro-1,4-benzothiazepine (K201, JTV519) with enhanced electron donor properties. Instead of activating channel activity (similar to 100 mu M), the 4-methoxy analog of 4-CmC [4-methoxy-3-methyl phenol (4-MmC)] inhibits channel activity at submicromolar concentrations (IC(50) = 0.34 +/- 0.08 mu M). Increasing the electron donor characteristics of K201 by synthesizing its dioxole congener results in an approximately 16 times more potent RyR1 inhibitor (IC(50) = 0.24 +/- 0.05 mu M) compared with K201 (IC(50) = 3.98 +/- 0.79 mu M). Inhibition is not caused by an increased closed time of the channel but seems to be caused by an open state block of RyR1. These alterations to chemical structure do not influence the ability of these drugs to affect Ca(2+)-dependent ATPase activity of sarcoplasmic/endoplasmic reticulum Ca(2+)-ATPase type 1. Moreover, the FKBP12 protein, which stabilizes RyR1 in a closed configuration, is shown to be a strong electron donor. It seems as if FKBP12, K201, its dioxole derivative, and 4-MmC inhibit RyR1 channel activity by virtue of their electron donor characteristics. These results embody strong evidence that designing new drugs to target RyR1 with enhanced electron donor characteristics results in more potent channel inhibitors. This is a novel approach to the design of new, more potent drugs with the aim of functionally modifying RyR1 single-channel activity.

Pd-Catalyzed Direct Arylations of Heteroarenes with Polyfluoroalkoxy-Substituted Bromobenzenes

The reactivity of di-, tri- and tetra-fluoroalkoxy-substituted bromobenzenes in the direct arylation of 5-membered ring heteroarenes using palladium catalysis was explored. High yields in arylated heteroarenes were obtained using only 1 mol-% of Pd(OAc)(2)catalyst with KOAc as an inexpensive base. Similar yields were obtained witho/m/ptrifluoromethoxy-,o/pdifluoromethoxy-, and tetrafluoroethoxy-substituents on the aryl bromide. A bromo-substituted difluorobenzo[d][1,3]dioxole was successfully coupled. The major side-products of the reaction are HBr/KOAc. Therefore, this synthetic scheme is very attractive for the access to such polyfluoroalkoxy-containing arylated heteroaromatics in terms of cost, simplicity, and low environmental impact, compared to reactions involving arylation of heteroarenes with bromophenols followed by polyfluoroalkylation.

Reverse osmosis separation of particular organic solvent mixtures by a perfluorodioxole copolymer membrane

Reverse osmosis (RO) separation of organic solvent mixtures where solvent molecular weights are < 100 Da is challenging especially for powerful solvents that swell most uncrosslinked polymers. To avoid polymer swelling by solvents, a particular perfluoropolymer, perfluoro-2,2-dimethyl-1,3-dioxole copolymerized with tetrafluoroethylene, (PDD-TFE), designated CMS-7, was studied. This amorphous glassy extremely hydrophobic copolymer has a very high free volume (FV) fraction. The maximum dimension of FV regions is less than similar to 0.65 nm allowing only single solvent molecule permeation. Further, interactions between polarity, dimensions and shapes of solvent molecules with those of polymer FV elements can lead to extraordinarily selective permeation. RO separation of the following binary systems through a thin 1.67 mu m film of this polymer supported on an e-PTFE support was studied over 1000-3500 kPa feed pressure: N-Methyl-2-pyrrolidone (NMP)-toluene; dimethylformamide (DMF)-toluene; dimethylsulfoxide (DMSO)-toluene; NMP-methanol; n-butanol-ethanol. Pure toluene appeared as permeate for three polar aprotic-aromatic systems from highly toluene-rich feeds; the membrane rejected polar molecules having dimensions similar to those of toluene. High osmotic pressure of the feed mixtures employed vis-a-vis feed pressures used reduced toluene permeation flux and the range of separable feed mixtures. Pure methanol permeate was obtained from particular polar aprotic-polar protic NMP-methanol mixtures due to significantly smaller methanol dimensions. Pure ethanol permeate was also similarly obtained from a particular mixture with n-butanol, a case of polar protic1-polar protic2 system where ethanol molecules were smaller. Such novel and clean-cut pressure-driven separations may be due to various combinations of the dimensions of the solvent molecules and the polymer FV regions, high polymer hydrophobicity, polarity of the aprotic/protic solvents and very low swelling of the polymer. Solvent sorption studies of dense polymers provide an insight into swelling of the polymer by the solvent and potential solvent-specific interactions. Sorption studies using 25 mu m polymer samples for polar aprotic solvents, NMP, DMF, DMSO, yielded very low levels of sorption, 0.4-0.9 wt%; those for toluene and methanol were already known to be only 1.34 and 1.2 wt% respectively. Permeation behaviors of other solvent mixtures, toluene-n-heptane, NMP-tetrahydrofuran, methanol-water, ethanol-water as well as individual phases of the immiscible mixture of NMP and the nonpolar solvent n-heptane were also studied. Ungeremine and Its Hemisynthesized Analogues as Bactericides against Flavobacterium columnare

The Gram-negative bacterium Flavobacterium columnare is the cause of columnaris disease, which can occur in channel catfish (Ictalurus punctatus). In a previous study, the betaine-type alkaloid ungeremine, 1, obtained from Pancratium maritimum L. was found to have strong antibacterial activity against F. columnare. In this study, analogues of 1 were evaluated using a rapid bioassay for activity against F. columnare to determine if the analogues might provide greater antibacterial activity and to determine structure-activity relationships of the test compounds. Several ungeremine analogues were prepared by hydrochlorination of the alkaloid and by selenium dioxide oxidation of both lycorine, 7, and pseudolycorine, 8, which yielded the isomer of ungeremine, 3, and zefbetaine, 4, respectively. The treatment of lycorine with phosphorus oxychloride allowed the synthesis of an anhydrolycorine lactam, 5, showing, with respect to 1, the deoxygenation and oxygenation of C-2 and C-7 of the C and B rings, respectively. The results of the structure-activity relationship studies showed that the aromatization of the C ring and the oxidation to an azomethine group of C-7 of the B ring are structural features important for antibacterial activity. In addition, the position of the oxygenation of the C ring as well as the presence of the 1,3-dioxole ring joined to the A ring of the pyrrolo[de]phenanthridine skeleton also plays a significant role in imparting antibacterial activity. On the basis of 24-h 50% inhibition concentration (IC50) results, ungeremine hydrochloride, 2, was similar in toxicity to 1, whereas 5 had the lowest activity. Analogue 2 is soluble in water, which may provide the benefit for use as an effective feed additive or therapeutant compared to ungeremine.

Catechols are components of many metal-chelating compounds, including siderophores that are naturally occurring iron(III) chelators excreted by microorganisms. The catechol can subsequently be deprotected, at the end of the synthesis, with trifluoroacetic acid (TFA). This strategy was applied to the synthesis of two catechol compounds functionalized with a terminal propargyl extension. These two compounds were shown to promote iron uptake in Escherichia coli and Pseudomonas aeruginosa. These two compounds are suitable for use as vectors in antibiotic Trojan horse approaches, as they could be conjugated with azide-functionalized antibiotics using the Huisgen dipolar 1,3-cycloaddition.
