Palmatine and berberine, structurally similar isoquinoline alkaloids exhibiting a broad range of biological activities, were recently found to inhibit p300 histone acetyltransferase (HAT), a potential therapeutic target for treating transcriptional activator-driven malignancies and diseases. Here, we report the first total synthesis of B-homo palmatine (11a) and berberine (11b) derivatives, which were synthesized from 3,4-dimethoxybenzaldehyde (1a) and benzo[d][1,3]dioxole-5-carbaldehyde (1b) in nine steps in 13.8 and 16.9% overall yields, respectively. A number of other new B-homo palmatine and berberine derivatives were also prepared. These derivatives display good inhibitory activity against p300 HAT; compound 12a manifests the most potent inhibition with an IC50 value of 0.42 mu m. Cell-based assays revealed that 12a exhibits certain inhibitory activity against HCG27, HT1080, and Z-138 cell lines, and no visible activity towards other cancer cell lines tested, reflecting that 12a has low cytotoxicity and acts against some types of cancer cells.

A theoretical analysis on planar and puckered-ring conformations of cyclopentadiene, 2,3-dihydrofuran, and 1,3-dioxole is carried out to test the conclusion of Laane and co-workers (J. Am. Chem. Soc. 1993, 115, 12132) that the unexpected nonplanarity of 1,3-dioxole must be attributed to the anomeric effect. MP4/6-31G**/IMP2/6-31G** calculations and NBO analysis show that delocalization involving the oxygen lone pairs and the C-O antibonding orbital, commonly associated with the anomeric effect, plays a decisive role in explaining the above-mentioned experimental fact. A careful analysis of the dipole-dipole interaction energy indicates that, in the present case, the electrostatic theory does not help to rationalize the experimentally observed puckered-ring conformation of 1,3-dioxole.

These copolymers are thermally stable (the initial decomposition temperatures are higher than 350 degrees C under an N-2 atmosphere) and have low refractive indices and high optical transparency from UV to near-infrared. Regulating the fluid flow in microfluidic devices enables a wide range of assay protocols for analytical applications. A programmable, photo-paper-based microfluidic device fabricated by using a method of cutting and laminating, followed by printing, is reported. The flow distance of fluid in the photo-paper-based channel was linearly proportional to time. By printing silver nanoparticle (AgNP) and poly.4,5-difluoro-2,2-bis.trifluoromethyl)-1,3-dioxole-co-tetrafluoroethylene] (PTFE) patterns on the surface of a photo-paper-based channel, we were able to either increase or decrease the fluid flow in the fabricated microfluidic devices, while maintaining the linearity in the flow distance-time relation. In comparison to the speed of fluid flow in a pristine channel, by using hydrophilic AgNP patterns, we were able to increase the speed in the channel by up to 15 times while we were able to slow the speed by a factor of 3 when using hydrophobic PTFE dots. We then further demonstrated a single-step protocol for detecting glucose and a multi-step protocol for detecting methyl paraoxon (MPO) with our methods in photo-paper-based microfluidic devices. This approach can lead to improved fluid handling techniques to achieve a wide range of complex, but programmable, assays without the need for any additional auxiliary devices for automated operation.

Thermal, photosynthesis and antibacterial studies of bioactive safrole derivative as precursor for natural flavor and fragrance

Safrole [5-allylbenzo[d][1,3]dioxole] was subjected to photochemical oxidation reaction with hydrogen peroxide in the presence of sodium lamp to give the corresponding epoxy derivative [5-oxiranylmethylbenzo[1,3]dioxole. The thermal oxidation of safrole with 3-chloroperoxybenzoic acid at room temperature gave the same epoxide derivative in quantitative yield. Antibacterial studies were carried out on safrole and its photoproducts ( safrole epoxide and safrole hydroperoxide). The results revealed that safrole hydroperoxide was the most effective than safrole epoxide than safrole against Gram-positive bacteria Bacillus subtilis ATCC6633, Staphylococcus aureus ATC C25923, and Gram negative bacteria Escherichia coli ATCC25422. Preparation of Hyflon AD60/PVDF composite hollow fiber membranes for vacuum membrane distillation

Hydrophobic membrane is desired to avoid pore wetting phenomenon in membrane distillation (MD). In the present paper, highly hydrophobic Hyflon AD60/PVDF composite hollow fiber membranes were prepared by coating a copolymer (Hyflon AD60) of tetrafluoroethylene (TFE) and 2,2,4-trifluoro-5-trifluoro methoxy-1,3-dioxole (TTD) on poly(vinylidene fluoride) (PVDF) membranes. The effects of Hyflon AD60 concentration, coating time, heat-treatment temperature and heat-treatment time on the structures and performances of Hyflon AD60/PVDF composite hollow fiber membranes in vacuum membrane distillation (VMD) process were investigated. The results show that the hydrophobicity of PVDF hollow fiber membrane was significantly improved by coating Hyflon AD60. The optimized Hyflon AD60 concentration, coating time, heat-treatment temperature and heat-treatment time were 0.1 wt%, 10-20 min, 40-50 degrees C and 9 h, respectively. Employing NaCl solution with concentration of 35 g x L-1, under a vacuum of 0.09 MPa and feed temperature of 70 degrees C, the water flux and salt rejection of the composite hollow fiber membrane in VMD process reached to approximately 10 kg x m(-2) x h(-1) and higher than 99.9 %, respectively, and kept stable, indicating that Hyflon AD60/PVDF composite membrane is an excellent candidate for operation in VMD operation. How does substitution affect the unimolecular reaction rates of Criegee intermediates?

To gain an understanding of the substitution effect on the unimolecular reaction rate coefficients for Criegee intermediates (CIs), we performed ab initio calculations for CH2OO, CH3CHOO, (CH3)(2)COO, CH3CH2CHOO, CH2CHCHOO and CHCCHOO. The energies of the CIs, products and transition states were calculated with QCISD(T)/CBS//B3LYP/6-311+ G(2d, 2p), while the rate coefficients were calculated with anharmonic vibrational correction by using second order vibrational perturbation theory. It was found that for single bonded substitutions, the hydrogen transfer reaction dominates for the syn-conformers, while the OO bending reaction dominates for the anti-conformers. However once a double bond or a triple bond is added, the OO bending reaction dominates for both syn and anti-conformers. The rate coefficients for OO bending reaction show a significant increase when adding a methyl group or ethyl group. On the other hand, the addition of unsaturated vinyl and acetylene groups usually results in a slower thermal decomposition compared to the substitution with saturated carbon groups. Interestingly, for syn_Syn-CH2CHCHOO, a special five member ring closure reaction forming dioxole was calculated to have an extremely fast rate coefficient of 9312 s(-1) at room temperature.
