Synthesis and characterization of mixed-ligand diimine-piperonal thiosemicarbazone complexes of ruthenium(II): Biophysical investigations and biological evaluation as anticancer and antibacterial agents

We have used a novel microwave-assisted method developed in our laboratories to synthesize a series of ruthenium-thiosemicarbazone complexes. The new thiosemicarbazone ligands are derived from benzo[d][1,3]dioxole-5-carbaldehyde (piperonal) and the complexes are formulated as [(diimine)(2) Ru(TSC)](PF6)(2) (where the TSC is the bidentate thiosemicarbazone ligand). The diimine in the complexes is either 2,2′-bipyridine or 1,10-phenanthroline. The complexes have been characterized by spectroscopic means (NMR, IR and UV-Vis) as well as by elemental analysis. We have studied the biophysical characteristics of the complexes by investigating their anti-oxidant ability as well as their ability to disrupt the function of the human topoisomerase ll enzyme. The complexes are moderately strong binders of DNA with binding constants of 10(4) M-1. They are also strong binders of human serum albumin having binding constants on the order of 10(4) M-1. The complexes show good in vitro anticancer activity against human colon cancer cells, Caco-2 and HCT-116 and indeed show some cytotoxic selectivity for cancer cells. The IC50 values range from 7 to 159 mu M (after 72 h drug incubation). They also have antibacterial activity against Gram-positive strains of pathogenic bacteria with IC50 values as low as 10 mu M: little activity was seen against Gram-negative strains. New chlorinated amphetamine-type-stimulants disinfection-by-products formed during drinking water treatment

Previous studies have demonstrated high removal rates of amphetamine-type-stimulants (ATSs) through conventional drinking water treatments; however the behaviour of these compounds through disinfection steps and their transformation into disinfection-by-products (DBPs) is still ‘unknown. In this work, for the first time, the reactivity of some ATSs such as amphetamine, methamphetamine, 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxymethamphetamine (MDMA) and 3,4-methylenedioxyethylamphetamine (MDEA) with chlorine has been investigated under simulated and real drinking water treatment conditions in order to evaluate their ability to give rise to transforrnation products. Two new DBPs from these illicit drugs have been found. A common chlorinated-by-product (3-chlorobenzo)-1,3-dioxole, was identified for both MDA and MDEA while for MDMA, 3-chlorocatechol was found. The presence of these DBPs in water samples collected through drinking water treatment was studied in order to evaluate their formation under real conditions. Both compounds were generated through treatment from raw river water samples containing ATSs at concentration levels ranging from 1 to 15 ng/L for MDA and from 2.3 to 78 ng/L for MDMA. One of them, (3-chlorobenzo)-1,3-dioxole, found after the first chlorination step, was eliminated after ozone and GAC treatment while the MDMA DBP mainly generated after the postchlorination step, showed to be recalcitrant and it was found in final treated waters at concentrations ranging from 0.5 to 5.8 ng/L. Discovery and Mechanistic Study of a Novel Human-Stimulator-of-Interferon-Genes Agonist

Stimulator of interferon genes (STING) is an integral ER-membrane protein that can be activated by 2’3′-cGAMP synthesized by cyclic guanosine monophosphate-adenosine monophosphate synthase (cGAS) upon binding of double-stranded DNA. It activates interferon (IFN) and inflammatory cytokine responses to defend against infection by microorganisms. Pharmacologic activation of STING has been demonstrated to induce an antiviral state and boost antitumor immunity. We previously reported a cell-based high-throughput-screening assay that allowed for identification of small-molecule cGAS-STING-pathway agonists. We report herein a compound, 6-bromo-N-(naphthalen-1-yl)-benzo[d][1,3]dioxole-5-carboxamide (BNBC), that induces a proinflammatory cytokine response in a human-STING-dependent manner. Specifically, we showed that BNBC induced type I and III IFN dominant cytokine responses in primary human fibroblasts and peripheral-blood mononuclear cells (PBMCs). BNBC also induced cytokine response in PBMC-derived myeloid dendritic cells and promoted their maturation, suggesting that STING-agonist treatment could potentially regulate the activation of CD4+ and CD8+ T lymphocytes. As anticipated, treatment of primary human fibroblast cells with BNBC induced an antiviral state that inhibited the infection of several kinds of flaviviruses. Taken together, our results indicate that BNBC is a human-STING agonist that not only induces innate antiviral immunity against a broad spectrum of viruses but may also stimulate the activation of adaptive immune responses, which is important for the treatment of chronic viral infections and tumors.

Copolymerization of tetrafluoroethylene and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole in Supercritical carbon dioxide

Copolymers of tetrafluoroethylene (TFE) and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole (PDD) were synthesized in carbon dioxide at low temperatures. Bis(perfluoro-2-N-propoxypropionyl) peroxide was used as initiator. A range of copolymers with various compositions and different molecular weights were prepared in yields as high as 74%. The glass transition temperature of the copolymers increased with increasing PDD content and ranged from 67 to 334degreesC for the PDD homopolymer. The phase behavior was found to depend on the feed composition. A comparison of a fully fluorinated commercially available product with a sample synthesized in carbon dioxide (without post-fluorination procedures) to match the composition and the molecular weight of the commercial sample showed no significant difference using IR spectroscopy, NMR spectroscopy, and differential scanning calorimetry, indicating similar microstructures. A simple method to determine the copolymer composition based on infrared spectroscopy was developed.

Chain structure and stiffness of Teflon AF glassy amorphous fluoropolymers

The structure of the amorphous perfluorinated polymer Teflon AF 2400 and other structurally close perfluoropolymers was studied by means of a quantum chemistry method. The electronic and structural characteristics of the repeating unit and polymer models with ten and nine monomer units were obtained. It was found that two nonplanar isomers can exist for different models of the perfluorinated dioxole ring with a difference of their energy minimums of 10.8 kJ/mol. The orthogonal-block structure of the polymer chain of the perfluorodioxole homopolymer and its copolymer with tetrafluoroethylene was proposed, the block size was found, and a possible diameter of the void formed by two neighboring polymer chains was evaluated. Potential energy curves for the rotation of certain chain fragments about different bonds of the polymer main chain were constructed, and the polymer stiffness was shown to substantially depend on the molar ratio between perfluorodioxole and tetrafluoroethylene units in the copolymer and on the geometry of the perfluorodioxole ring.

However, in addition, 3,4-dichloro-7-methoxy-5-(4-methoxyphenyl)-5,10-dihydrophenazine-1,2-dione was formed in the reaction of 1-(4-methoxyphenyl)-2-(4-methoxyphenylaminomethylene)-4,5 -diphenyl- 1,2-dihydropyrrol-3-one with tetrachloro-1,2-benzoquinone.

Perfluoro-4-methyl-1,3-dioxole: a new monomer for high-T-g amorphous fluoropolymers

A 4-Chloro-5-trifluoromethyl-2,2,4-trifluoro-1,3-dioxolane (1) was synthesised by reaction of CF2(OF)(2) with CF3-CH=CFCl; the elimination of HCl from (1) in basic conditions led to the formation of dioxole perfluoro-4-methyl-1,3-dioxole (2). Treatment of 1 with Au(I) catalyst in wet CH2Cl2 produced either 2 or 3 as a major product depending on the temperature, reaction time. and catalyst loading. D has been proposed as an intermediate, which might be formed via Au(I)-induced internal oxacyclization of the intermediate 4 followed by chemoselective nucleophilic attack by the water molecule. Formation of 2 or 3 might be explained via sequential 1,3-dioxole ring opening and gold-promoted 5-endo-dig carbocyclization or simple protonation of the intermediate D, respectively.

Accurate masses of these impurities were determined by using a Q-TOF mass spectrometer. Based on chromatographic, spectrometric data and plausible chemical transformation mechanism, the structures of IMP-I, IMP-II, IMP-III and IMP-IV were identified, respectively, as bifendate, (E)-5-(2,4-ditert- butyl-6-((2,4-dioxothiazolidin-5-ylidene) methyl) phenyl)-5′-methyl-7,7′- dimethoxy-[4,4′-bibenzo[d][1,3] dioxole]-5,5′-dicarboxylate, (Z)-5( 3,5-di-tert-butyl-2-hydroxybenzylidene) thiazolidine-2,4-dione and bis(2,4-di-tert-butyl-6-((Z)-(2,4-dioxothiazolidin-5-ylidene) methyl)phenyl)- 7,7′-dimethoxy-[4,4′-bibenzo[d][1,3] dioxole]-5,5′-icarboxylate.
