Comparison of the oxidation of carcinogenic aristolochic acid I and II by microsomal cytochromes P450 in vitro: experimental and theoretical approaches

The herbal drug aristolochic acid, a natural mixture of 8-methoxy-6-nitrophenanthro[3,4-d]-1,3-dioxole-5-carboxylic acid (AAI) and 6-nitrophenanthro[3,4-d]-1,3-dioxole-5-carboxylic acid (AAII), is derived from Aristolochia species and is the cause of two nephropathies. Ingestion of aristolochic acid is associated with the development of urothelial tumors linked with aristolochic acid nephropathy and is implicated in the development of Balkan endemic nephropathy-associated urothelial tumors. The O-demethylated metabolite of AAI, 8-hydroxyaristolochic acid (AAIa), is the detoxification product of AAI generated by its oxidative metabolism. Whereas the formation of AAIa from AAI by cytochrome P450 (CYP) enzymes has been found in vitro and in vivo, this metabolite has not been found from AAII as yet. Therefore, the present study has been designed to compare the amenability of AAI and AAII to oxidation; experimental and theoretical approaches were used for such a study. In the case of experimental approaches, the enzyme (CYP)-mediated formation of AAIa from both carcinogens was investigated using CYP enzymes present in subcellular microsomal fractions and recombinant CYP enzymes. We found that in contrast to AAI, AAII is oxidized only by several CYP enzymatic systems and their efficiency is much lower for oxidation of AAII than AAI. Using the theoretical approaches, such as flexible in silico docking methods and ab initio calculations, contribution to explanation of these differences was established. Indeed, the results found by both used approaches determined the reasons why AAI is better oxidized than AAII; the key factor causing the differences in AAI and AAII oxidation is their different amenability to chemical oxidation.

Vasodilator and antihypertensive effects of a novel N-acylhydrazone derivative mediated by the inhibition of L-type Ca2+ channels

New bioactive N-acylhydrazone derivatives synthesized from safrole previously have been found to promote intense vasodilation and antihypertensive activity. In this study, we describe the synthesis and the cardiovascular effects of the new N-acylhydrazone derivative (E)-N-methyl-N ‘-(thiophen-3-ylmethylene)benzo[d][1,3]dioxole-5-carbohydrazide (LASSBio-1289). Thoracic aorta and left papillary muscles from Wistar-Kyoto (WKY) rats and spontaneously hypertensive rats (SHR) were prepared for isometric tension recording. LASSBio-1289 promoted relaxation of endothelium-intact and denuded aortic rings with respective pIC(50) (-log IC50) values of 5.07 +/- 0.09 and 4.26 +/- 0.09 (P<0.001) for WKY rats and 5.43 +/- 0.05 and 5.58 +/- 0.07 (P>0.05) for SHR. The vasodilator activity of LASSBio-1289 was increased in the KCl-contracted aorta. LASSBio-1289 attenuated the contracture elicited by Ca2+ in depolarized aorta from both WKY rats and SHR. In endothelium-intact aorta from WKY rats, LASSBio-1289-induced relaxation was unchanged after incubation with propranolol, ZM 241385, atropine, diphenhydramine, and HOE140, but was significantly reduced by L-NAME and ODQ. LASSBio-1289 decreased papillary muscles contractility only at concentrations above 200 mu m. Acute intravenous injection of LASSBio-1289 (3mg/kg) produced a significant hypotensive response in SHR but not in WKY rats, suggesting its antihypertensive profile. The antihypertensive effect was also observed in SHR during 14days of intraperitoneal and oral administration. In conclusion, our data demonstrated that LASSBio-1289 induces both endothelium-independent vasorelaxation involving the inhibition of Ca2+ influx through L-type Ca2+ channels in aorta from WKY rats and SHR, and endothelium-dependent relaxation mediated by the NO/cyclic GMP pathway in WKY rats.

Experimental and Computational Evaluation of Piperonylic Acid Derived Hydrazones Bearing Isatin Moieties as Dual Inhibitors of Cholinesterases and Monoamine Oxidases

A set of piperonylic acid derived hydrazones with variable isatin moieties was synthesized and evaluated for their inhibitory activity against the enzymes acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and monoamine oxidases A and B (MAO-A/B). The results of in vitro studies revealed IC50 values in the micromolar range, with the majority of the compounds showing selectivity for the MAO-B isoform. N-[2-Oxo-1-(prop-2-ynyl)indolin-3-ylidene]benzo[d][1,3]dioxole-5-carbohydrazide (3) was identified as a lead AChE inhibitor with IC50=0.052 +/- 0.006 mu m. N-[(3E)-5-chloro-2-oxo-2,3-dihydro-1H-indol-3-ylidene]-2H-1,3-benzodioxole-5-carbohydrazide (2) was the lead MAO-B inhibitor with IC50=0.034 +/- 0.007 mu m, and showed 50 times greater selectivity for MAO-B over MAO-A. The kinetic studies revealed that compounds 2 and 3 displayed competitive and reversible inhibition of AChE and MAO-B, respectively. The molecular docking studies revealed the significance of hydrophobic interactions in the active site pocket of the enzymes under investigation. Further optimization studies might lead to the development of potential neurotherapeutic agents.

Interplay of structure and reactivity in a most unusual furan Diels-Alder reaction

Difluorinated alkenoate ethyl 3,3-difluoro-2-(N, N-diethylcarbamoyloxy)-2-propenoate reacts rapidly and in high yield with furan and a range of substituted furans in the presence of a tin(IV) catalyst. In the presence of both catalyst and solvent, a two-step reaction is predicted, corresponding to the stepwise formation of the two new carbon-carbon bonds via transition states which have similar energies in all cases. Our experimental observations of the lack of reaction of the non-fluorinated dienophile, the stereochemical outcomes, and the rate acceleration accompanying furan methylation are all well predicted by our calculations. The calculated free energy barriers generally correlate well with measured reaction rates, supporting a reaction mechanism in which zwitterionic character is developed strongly. An in situ ring opening reaction of exo-cycloadduct ethyl exo-2-(N, N-diethylcarbamoyloxy)-3,3-difluoro-7-oxabicyclo[2.2.1] hept-5-enyl-2-endo-carboxylate, which results in the formation of cyclic carbonate ethyl 4,4-difluoro-5-hydroxy-2-oxo-5,7a-dihydro-4H-benzo[1,3] dioxole-3a-carboxylate by a Curtin-Hammett mechanism, has also been examined. Substantial steric opposition to Lewis acid binding prevents carbonate formation from 2-substituted furans.

It has been previously reported(1-4) that standard 193 nm materials, like Teflon AF(R) and Cytop(R), fail catastrophically upon exposure at 157 nm radiation; and that their transmission is very poor to non-existent. In this paper we report data showing that these materials have higher transmission and lifetimes than previously thought 2. The physical lifetime of Cytop(R) of nearly 500 J/cm(2) is remarkably high, although the transmission varies as a function dose suggesting physical and chemical changes from the onset. For Teflon AF(R) the transmission is a more complex function of exposure dose, suggesting competing mechanisms. From experiments run with PVDF (polyvinyl difluoride), we conclude that the widely reported photodarkening effect is due to the presence of hydrogen in close proximity to a fluorine atom. From IR spectra we conclude that the dioxole moeity in Teflon AF(R) undergoes a series of photochemical reactions that lead to the physical destruction of the polymer.

Synthesis and Biological Activity of Some New N-bridged Heterocycles of Piperonal-based Triazolo-thiadiazoles

A new series of 6-(6-nitro/bromo/hydrobenzo[d][1,3]dioxol-5-yl)-3-alkyl/aryl-[1,2,4] triazolo[3,4-b][1,3,4]thiadiazoles (4a-o) was synthesized by cyclocondensation of 6-nitro/bromobenzo/hydro[d][1,3]dioxole-5-carboxylic acids (2a-c) with 3-alkyliary1-4-amino-5-mercapto-1,2,4-triazoles (3a-e) in the presence of phosphorus oxychloride as a cyclizing agent. The newly synthesized compounds were characterized by LC-MS, FT-IR, H-1-NMR, and also CHN analysis. All the newly synthesized compounds were tested for their bacterial inhibition ability along with antioxidant properties. Among the synthesized compounds, compounds 4o (11.0 +/- 1.4) and 4l (10.5 +/- 0.20) exhibited good bacterial inhibition against E. coli, whereas compounds 4i and 4g also showed (10.5 +/- 1.8) and (10.0 +/- 0.70) good inhibition against B. subtilis and S. aureus. respectively. The majority of the synthesized compounds exhibited poor antioxidant activity due to lack of electron/proton donor capacity in the synthesized compounds.
