Designing Calcium Release Channel Inhibitors with Enhanced Electron Donor Properties: Stabilizing the Closed State of Ryanodine Receptor Type 1

New drugs with enhanced electron donor properties that target the ryanodine receptor from skeletal muscle sarcoplasmic reticulum (RyR1) are shown to be potent inhibitors of single-channel activity. In this article, we synthesize derivatives of the channel activator 4-chloro-3-methyl phenol (4-CmC) and the 1,4-benzothiazepine channel inhibitor 4-[-3{1-(4-benzyl) piperidinyl}propionyl]-7-methoxy-2,3,4,5-tetrahydro-1,4-benzothiazepine (K201, JTV519) with enhanced electron donor properties. Instead of activating channel activity (similar to 100 mu M), the 4-methoxy analog of 4-CmC [4-methoxy-3-methyl phenol (4-MmC)] inhibits channel activity at submicromolar concentrations (IC(50) = 0.34 +/- 0.08 mu M). Increasing the electron donor characteristics of K201 by synthesizing its dioxole congener results in an approximately 16 times more potent RyR1 inhibitor (IC(50) = 0.24 +/- 0.05 mu M) compared with K201 (IC(50) = 3.98 +/- 0.79 mu M). Inhibition is not caused by an increased closed time of the channel but seems to be caused by an open state block of RyR1. These alterations to chemical structure do not influence the ability of these drugs to affect Ca(2+)-dependent ATPase activity of sarcoplasmic/endoplasmic reticulum Ca(2+)-ATPase type 1. Moreover, the FKBP12 protein, which stabilizes RyR1 in a closed configuration, is shown to be a strong electron donor. It seems as if FKBP12, K201, its dioxole derivative, and 4-MmC inhibit RyR1 channel activity by virtue of their electron donor characteristics. These results embody strong evidence that designing new drugs to target RyR1 with enhanced electron donor characteristics results in more potent channel inhibitors. This is a novel approach to the design of new, more potent drugs with the aim of functionally modifying RyR1 single-channel activity.

Benzodioxole derivatives as negative effectors of plant proteases

Previous work demonstrated that a commercial formulation of piperonyl butoxide (PBO) did inhibit the activity of some plant proteolytic enzymes. In this paper, the effect of pure PBO and nine pure PBO homologues (PBOH) appropriately synthesized toward bromelain and papain was studied in hydrocarbon solution using the bis(2-ethylhexyl)sodium sulfosuccinate (AOT) reverse micellar system. This study establishes that the majority of these compounds show, in vitro, interesting protease inhibition activities. The benzodioxole and dihydrobenzofuran structures, in particular, 5-[2-(2butoxyethoxy)ethoxymethyl]-benzo[1,3]dioxole (EN 1-40) and 6-[2-(2-butoxyethoxy)ethoxymethyl]5-propyl-2,3-dihydrobenzofuran (EN 16-5), respectively, appear to be responsible for protease inhibition. Measures of octanol/water partition coefficients on PBO and PBOH have demonstrated that water solubility plays a fundamental role in the expression of protease inhibition activity.

The role of lone pair and dipolar interactions in the non-planarity of 1,3-dioxolane and 1,3-dioxole

1,3-Dioxole has been shown to be non-planar by infrared and Raman spectroscopy. An MM3 study of this molecule enabled the investigators to suggest that this non-planarity was due to the anomeric effect. Subsequently, an ab initio theoretical study of this molecule was performed, which also concluded that the non-planarity of 1,3-dioxole was due to the anomeric effect and not to dipole-dipole interactions. Neither study used rigorous methods for assessing the role of dipolar interactions in the geometry of 1,3-dioxole. A new study of 1,3-dioxole, 1,3-dioxolane, tetrahydrofuran, cyclopentane, and some related molecules using the new QVBMM (molecular mechanics) force field shows conclusively that the non-planarity of 1,3-dioxole and 1,3-dioxolane is due primarily to torsional and dipolar effects, and not secondary molecular orbital overlap interactions.

EFFECT OF X-IRRADIATION ON OPTICAL-PROPERTIES OF TEFLON-AF

Radiation effects in optical-grade amorphous fluoropolymer, Teflon-AF, is investigated by UV-visible absorption and electron spin resonance (ESR) measurements. When irradiated with low-energy (40 kVp) X-rays at room temperature in air, Teflon-AF is found to develop a broad, structureless UV-absorption band in the wavelength interval 200-350 nm. While the UV absorption increases as a function of X-ray dose, with relative rates of approx 2 x 10(-5) Gy-1 (1 x 10(-5) Gy-1) in Teflon-AF 1600 (Teflon-AF 2400), its optical transparency for a given dose of 67.5 kGy, however, remains unaffected. Additional measurements conducted using electron spin resonance (ESR) technique reveal that the observed UV absorption is caused by the X-ray induced peroxy radical (POO.). The results also suggest that the inclusion of dioxole monomer in the PTFE chain not only improves the optical clarity of Teflon-AF, as reported, but also increases its radiation tolerance. During a post-irradiation storage in air at RT for about 30 days the peroxy radical is observed to decay, with a concomitant decrease in UV absorption. A tentative model is proposed to explain the radiation damage and recovery mechanisms.

Thermal desorption spectroscopy of PTFE dielectric films deposited on silicon wafers

Fluoropolymers are receiving considerable attention for use as integrated circuit dielectric materials. There is considerable confusion regarding the thermal stability of these materials. It is desirable to discuss thermal stability in terms of the outgassing properties of thin films deposited onto silicon wafers, since this is a key indicator of the fitness for use of a given dielectric film technology. In this paper we discuss the various types of fluoropolymer films and review previously published data on their thermal stability. Two such fluoropolymers, polytetrafluoroethylene (PTFE) and a copolymer of tetrafluoroethylene and 2,2-bistrifluoromethy-1-4,5-difluoro-1,3-dioxole (TFE-PDD), have been previously proposed for use as integrated circuit dielectrics. Thermal Desorption Spectroscopy results for thin films of these two polymers are presented. These results show that PTFE has much lower outgassing than does TFE-PDD.

Anti-fingerprints fluorinated coating for anodized titanium avoiding color alteration

In this study amorphous fluorinated coatings applied to anodized titanium surface have been investigated. A copolymer between tetrafluoroethylene and perfluoro-4-trifluoromethoxy-1,3-dioxole (AD60) and two perfluoropolyether containing ammonium phosphate (F10) or triethoxysilane (S10) functionalities have been tested. To estimate the color alteration of the anodized titanium surfaces due to the application of the coatings, spectrophotometric analyses have been made. Water and n-dodecane contact angles as well as apparent surface energy have been evaluated. Ellipsometry and atomic force microscopy data have been used to measure the thickness of the fluorinated coatings. A tailored mechanical preliminary test has also been explored to evaluate the adhesion of the coatings on the anodized titanium surface. The resistance to surface soiling with castor oil was also preliminarily investigated. The fluorinated coating tested on anodized titanium showed a low apparent surface energy and high chromatic aspect conservation, this is particularly evident for the titanium anodized coated with triethoxysilane functionalities fluoropolymers S10.

Chemical constituents from the mycelia of Antrodia cinnamomea
