O-17, C-13, and H-1 NMR studies of p-pi conjugation in 2-substituted 4-methylene-1,3-dioxolanes and 4-methyl-1,3-dioxoles

The O-17 NMR spectra of a number of unsaturated 5-membered cyclic acetals, 2-substituted 4-methylene- 1,3-dioxolanes and their endocyclic isomers, 4-methyl-1,3-dioxoles, have been recorded. The O-17 NMR chemical shifts, in comparison with those of similarly 2-substituted 1,3-dioxolanes, were used to explore the variation of the strength of p-pi conjugation in the unsaturated acetals as a function of the nature of substitution at C2. The O-17 NMR shift data reveal that alkoxy substituents have a significantly more favorable effect on the strength of p-pi conjugation in 4-methyl-1,3-dioxoles than in 4-methylene-1,3-dioxolanes. This fact appears to be responsible for the previously observed unexpectedly large effect of alkoxy substitution on the relative thermodynamic stabilities of these two classes of isomeric compounds. Additional information of the unexpected charge distribution in 4-methyl-1,3-dioxoles is provided by their H-1 and C-13 NMR spectra.

Novel furfurylidene N-acylhydrazones derived from natural safrole: discovery of LASSBio-1215, a new potent antiplatelet prototype

We describe herein the discovery of (E)-N-methyl-N’-((5-nitrofuran-2-yl) methylene) benzo[d][1,3]dioxole-5-carbohydrazide (9e), named LASSBio-1215, as a novel antiplatelet agent belonging to the N-methyl-N-acylhydrazone class, which exert their antiaggregating actions on human and rabbit platelets induced by different agonists, through cyclooxygenase-1 (COX-1) or thromboxane synthase inhibition. This compound was elected after screening of a series of functionalized furyl N-acylhydrazone derivatives, synthesized from natural safrole 10. In vitro assays showed that compound 9e presents platelet-aggregating activity in rabbit platelet-rich plasma (PRP) induced by arachidonic acid (IC50 = 0.7 mu M) and collagen (IC50 = 4.5 mu M). Moreover, LASSBio-1215 also inhibited almost completely the second wave of adenosine diphosphate-induced platelet aggregation in human PRP, and this effect was correlated with their ability to block the production of pro-aggregating autacoid thromboxane A(2).

Human Formalin-Fixed Paraffin-Embedded Tissues: An Untapped Specimen for Biomonitoring of Carcinogen DNA Adducts by Mass Spectrometry

DNA adducts represent internal dosimeters to measure exposure to environmental and endogenous genotoxicants. Unfortunately, in molecular epidemiologic studies, measurements of DNA adducts often are precluded by the unavailability of fresh tissue. In contrast, formalin-fixed paraffin embedded (FFPE) tissues frequently are accessible for biomarker discovery. We report here that DNA adducts of aristolochic acids (AAs) can be measured in FFPE tissues at a level of sensitivity comparable to freshly frozen tissue. AAs are nephrotoxic and carcinogenic compounds found in Aristolochia herbaceous plants, many of which have been used worldwide for medicinal purposes. AAs are implicated in the etiology of aristolochic acid nephropathy and upper urinary tract carcinoma. 8-Methoxy-6-nitrophenanthro-[3,4-d]-1,3-dioxole-5-carboxylic acid (AA-I) is a component of Aristolochia herbs and a potent human urothelial carcinogen. AA-I reacts with DNA to form the aristolactam (AL-I)-DNA adduct 7-(deoxyadenosin-N-6-yl) aristolactam I (dA-AL-I). We established a method to quantitatively retrieve dA-AL-I from FFPE tissue. Adducts were measured, using ultraperfonnance liquid chromatography/mass spectrometry, in liver and kidney tissues of mice exposed to AA-I, at doses ranging from 0.001 to 1 mg/kg body weight. dA-AL-I was then measured in 10-mu m thick tissue-sections of FFPE kidney from patients with upper urinary tract cancers; the values were comparable to those observed in fresh frozen samples. The limit of quantification of dA-AL-I was 3 adducts per 109 DNA bases per 2.5 mu g of DNA. The ability to retrospectively analyze FFPE tissues for DNA adducts may provide clues to the origin of human cancers for which an environmental cause is suspected.

Novel amorphous perfluorocopolymeric system: Copolymers of perfluoro-2-methylene-1,3-dioxolane derivatives

Perfluorotetrahydro-2-methylene-furo[3,4-d][1,3]dioxole (monomer I) and perfluoro-2-methylene-4-methoxymethyl-1,3-dioxolane (monomer II) are soluble in perfluorinated or partially fluorinated solvents and readily polymerize in solution or in bulk when initiated by a free-radical initiator, perfluorodibenzoyl peroxide. The copolymerization parameters have been determined with in situ F-19 NMR measurements. The copolymerization reactivity ratios are r(I) = 1.80 and r(II) = 0.80 in 1,1,2-trichlorotrifluoroethane at 41 degrees C and r(I) = 0.97 and r(II) = 0.85 for the bulk polymerization. These data show that this copolymerization pair has a good copolymerization tendency and yields nearly ideal random copolymers. The copolymers have only one glass-transition temperature from 101 to 168 degrees C, depending on the copolymer compositions. Melting endotherms have not been observed in their differential scanning calorimetry traces, and this indicates that all the copolymers with different compositions are completely amorphous. These copolymers are thermally stable (the initial decomposition temperatures are higher than 350 degrees C under an N-2 atmosphere) and have low refractive indices and high optical transparency from UV to near-infrared. Copolymer films prepared by casting were flexible and tough. These properties make the copolymers ideal candidates as optical and electrical materials. (c) 2006 Wiley Periodicals, Inc.

Block copolymer compatibilized polymer: fullerene blend morphology and properties

Recent studies have shown the role of block copolymer as compatibilizer in tuning the phase separated morphology of the active layer so as to improve the overall photovoltaic efficiency of organic photovoltaic (OPV) devices. Here, we substantiate this observation by investigating the role of a rod-coil block copolymer poly-(3-hexylthiophene)-b-polystyrene (P3HT-b-PS) as compatibilizer in influencing the blend morphology and device performance of several polymer:fullerene blend systems. Fullerene derivatives N-(3-methoxypropyl)-2-(carboxyethyl)-5-(4-cyanophenyl) fulleropyrrolidine (NCPF) and N-(3-methoxypropyl)-2-(carboxyethyl)-5-(5, 5-difluorobenzo-dioxole) fulleropyrrolidine (FFNCPF) were synthesized using Prato reaction, while P3HT-b-PS copolymer was synthesized using the combination of Grignard metathesis, ATRP, and click chemistry. The addition of P3HT-b-PS in P3HT:PCBM blend led to the formation of more homogenous structure compared to the pure P3HT:PCBM blend. Also, a reduction in domain size was observed in P3HT: FFNCPF system upon P3HT-b-PS addition which can be attributed to the compatibilization effect of BCP. Incorporation of P3HT-b-PS block copolymer was found to effectively alter the thin film nanostructure of polymer/fullerene derivative blends and polymer crystalline structure. Maximum enhancement in power conversion efficiency (PCE) of compatabilized P3HT:PCBM blend system was noticed followed by P3HT:FFNCPF and P3HT:NCPF. The moderate improvement in photovoltaic properties can be correlated with face-on orientation of P3HT crystallites and the segregation of the fullerene domains at the bulk heterojunction (BHJ) cathode interface which facilitates the efficient charge collection as respective electrodes. Sorption of gases and vapors in an amorphous glassy perfluorodioxole copolymer

Gas and vapor sorption properties of a random copolymer of 87 mol % 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole and 13% tetrafluoroethylene (AF2400) are reported. Using both pressure decay and inverse gas chromatography (IGC) methods, a wide range of solutes was studied: He, Nz, Oz, CO2, C-1-C-13 n-alkanes, CF4, C2F6, C6F6, and C6F5CF3. These solutes have critical temperatures ranging from 5 to 677 K. AF2400 has very large solubility coefficients, S, relative to other glassy and rubbery polymers. Only poly(1-trimethylsilyl-1-propyne), the most permeable polymer known, exhibits higher solubility coefficients. The large solubility coefficients in AF2400 are mainly due to high Henry’s law solubility coefficients. Fluorocarbon solutes exhibit higher solubility than their hydrocarbon analogues. A novel linear correlation between the logarithm of S and T-c(2), where T-c is the solute critical temperature, was observed. On the basis of IGC results, the microcavity size in this perfluoropolymer is larger than in conventional hydrocarbon-based glassy polymers.

Synthesis and polymerization of a novel perfluorinated monomer

Perfluoro(5-methylene-2,2-dimethyl-1,3-dioxolane) (1) was synthesized by utilizing a direct fluorination reaction. Compound 1 was an entirely novel monomer with difluoromethylene at position 5 on the dioxolane ring as an unprecedented polymerization site. It successfully polymerized with tetrafluoroethylene to afford copolymers, which had T-g values in the range of 60-90 degrees C. The content of monomer 1 in the obtained polymers was less than 20 mol%, which seemed insufficient for giving various unique properties to polymers. However, each polymer was expected to be a superior material because of their advanced thermal stability.
