Carbon dioxide (CO2) plasticization and sorption effects in both thick and thin films of high free-volume glassy perfluoropolymers were studied by monitoring CO2 permeability and by observing changes in the film thickness and refractive index with ellipsometry measurements. The film thickness, aging time, thermal history and CO2 exposure protocols have significant effect on the absolute CO2 permeability and plasticization behavior of both thick and thin films. The extent of CO2 plasticization increases as film thickness decreases and as the aging time is increased. The as-cast films showed higher plasticization compared to films which were annealed above T-g; however, the CO2 permeability of both the ascast and annealed films continuously decreased during the depressurization step unlike other glassy polymers. In general, the various CO2 exposure protocols revealed lower CO2 plasticization for perfluoropolymers compared to other reported glassy polymers. The extent of CO2 sorption obtained from the ellipsometry measurements was found to decrease with the decrease in the excess volume and increase in the aging time for perfluoropolymers; in addition, the structural differences among the various glassy polymers resulting in different polymer-gas interactions also affects the overall sorption characteristics. The lower plasticization in perfluoropolymers compared to Matrimid was also confirmed from the smaller percent increase observed for the experimental diffusion coefficient compared to the theoretically predicted diffusion coefficient from the dual sorption-mobility model. The Langmuir sorption parameter, C-H’, and solubility at infinite dilution, So, obtained from fitting dual sorption-mobility model to sorption data, showed an excellent linear correlation with (T-g-35) degrees C. The CO2 diffusivity and permeability data obtained for thin films of various glassy polymers also showed a strong correlation with free volume. The somewhat unusual behavior of thin films of AF 2400 in comparison to other glassy polymers studied to date is believed to be related to the low cohesive energy density expected of perfluorinated structures and its high free volume resulting from the bulky dioxole comonomer. The BDMMBSH derivatives were characterized using different spectroscopic techniques, such as H-1-NMR, C-13-NMR, FTIR, and UV-Vis spectroscopy, and their crystal structures were analyzed using the single crystal X-ray diffraction method (SCXRDM). Subsequently, the BDMMBSH compounds were used for the significant detection of the carcinogenic heavy metal ion, lead (Pb2+), via a reliable electrochemical approach. A sensitive and selective Pb2+ sensor was developed via the deposition of a thin layer of BDMMBSH on a GCE with the conducting polymer matrix Nafion (NF). The sensitivity, LOQ, and LOD of the proposed sensor towards Pb2+ were calculated from the calibration curves to be 2220.0 pA mu M-1 cm(-2), 320.0 mM, and 96.0 pM, respectively. The validation of the BDMMBSH/GCE/NF sensor probe was performed via the selective determination of Pb2+ in spiked natural samples with a satisfactory and rational outcome.

The kinetics of entry into and exit from the cavity of cucurbit[7]uril (CB7) was studied by the stopped-flow method in water at various temperatures using pharmaceutically important natural isoquinoline alkaloids as guest compounds. Solvent swelling of polymers is of major importance in organic solvent nanofiltration (OSN) affecting membrane rejection of 200-1000 + Da solutes. A selected perfluoro type amorphous glassy copolymer, perfluoro-2,2-dimethyl-1,3-dioxole copolymerized with tetrafluoroethylene (PDD-TFE), is completely insoluble in all solvents except perflourinated solvents; further it showed very little swelling in solvents that are not of the perfluorinated type. Pure component permeation flux for a particular variety of this copolymer, CMS-7, was measured with a wide variety of solvents for two membrane thicknesses, 1.67 in and 0.6 pm, in a composite form supported on a porous ePTFE substrate. The feed pressure range was 1000-3000 kPa. The solvents included methanol, ethanol, ethyl acetate, n-heptane, tetrahydrofuran (THF), toluene, dimethylsulfoxide (DMSO); n-heptane showed much higher flux than the rest. Successful OSN was demonstrated with high solute rejection in methanol solutions of two dye solutes Safranin 0 (MW, 351 Da) and Brilliant Blue R (MW, 826 Da). Rejection for Safranin 0 varied between 94% and 97% depending on the pressure and membrane thickness; that for Brilliant Blue R varied in the range of 97-99.8%. Successful OSN was also demonstrated with Safranin 0 in THF with solute rejection varying between 94% and 97%. Permeation behaviors of various mixtures of two solvents, toluene and n-heptane, were studied. The measured values of very limited extent of sorption of a variety of organic solvents in 25 pm thick dense samples of this and a related perfluoro type copolymer, CMS-3, were employed to explain the observed behavior.

Exposure to the plant nephrotoxin and carcinogen aristolochic acid (AA) leads to the development of AA nephropathy, Balkan endemic nephropathy (BEN) and upper urothelial carcinoma (UUC) in humans. Beside AA, exposure to ochratoxin A (OTA) was linked to BEN. Although OTA was rejected as a factor for BEN/UUC, there is still no information whether the development of AA-induced BEN/UUC is influenced by OTA exposure. Therefore, we studied the influence of OTA on the genotoxicity of AA (AA-DNA adduct formation) in vivo. AA-DNA adducts were formed in liver and kidney of rats treated with AA or AA combined with OTA, but no OTA-related DNA adducts were detectable in rats treated with OTA alone or OTA combined with AA. Compared to rats treated with AA alone, AA-DNA adduct levels were 5.4- and 1.6-fold higher in liver and kidney, respectively, of rats treated with AA combined with OTA. Although AA and OTA induced NAD(P)H:quinone oxidoreductase (NQO1) activating AA to DNA adducts, their combined treatment did not lead to either higher NQO1 enzyme activity or higher AA-DNA adduct levels in ex vivo incubations. Oxidation of AA I (8-methoxy-6-nitrophenanthro[3,4-d]-1,3-dioxole-5-carboxylic acid) to its detoxification metabolite, 8-hydroxyaristolochic acid, was lower in microsomes from rats treated with AA and OTA, and this was paralleled by lower activities of cytochromes P450 1A1/2 and/or 2C11 in these microsomes. Our results indicate that a decrease in AA detoxification after combined exposure to AA and OTA leads to an increase in AA-DNA adduct formation in liver and kidney of rats.
