Copolymerization of tetrafluoroethylene and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole in Supercritical carbon dioxide

Copolymers of tetrafluoroethylene (TFE) and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole (PDD) were synthesized in carbon dioxide at low temperatures. Bis(perfluoro-2-N-propoxypropionyl) peroxide was used as initiator. A range of copolymers with various compositions and different molecular weights were prepared in yields as high as 74%. The glass transition temperature of the copolymers increased with increasing PDD content and ranged from 67 to 334degreesC for the PDD homopolymer. The phase behavior was found to depend on the feed composition. A comparison of a fully fluorinated commercially available product with a sample synthesized in carbon dioxide (without post-fluorination procedures) to match the composition and the molecular weight of the commercial sample showed no significant difference using IR spectroscopy, NMR spectroscopy, and differential scanning calorimetry, indicating similar microstructures. A simple method to determine the copolymer composition based on infrared spectroscopy was developed.

Discovery and Mechanistic Study of a Novel Human-Stimulator-of-Interferon-Genes Agonist

Stimulator of interferon genes (STING) is an integral ER-membrane protein that can be activated by 2’3′-cGAMP synthesized by cyclic guanosine monophosphate-adenosine monophosphate synthase (cGAS) upon binding of double-stranded DNA. It activates interferon (IFN) and inflammatory cytokine responses to defend against infection by microorganisms. Pharmacologic activation of STING has been demonstrated to induce an antiviral state and boost antitumor immunity. We previously reported a cell-based high-throughput-screening assay that allowed for identification of small-molecule cGAS-STING-pathway agonists. We report herein a compound, 6-bromo-N-(naphthalen-1-yl)-benzo[d][1,3]dioxole-5-carboxamide (BNBC), that induces a proinflammatory cytokine response in a human-STING-dependent manner. Specifically, we showed that BNBC induced type I and III IFN dominant cytokine responses in primary human fibroblasts and peripheral-blood mononuclear cells (PBMCs). BNBC also induced cytokine response in PBMC-derived myeloid dendritic cells and promoted their maturation, suggesting that STING-agonist treatment could potentially regulate the activation of CD4+ and CD8+ T lymphocytes. As anticipated, treatment of primary human fibroblast cells with BNBC induced an antiviral state that inhibited the infection of several kinds of flaviviruses. Taken together, our results indicate that BNBC is a human-STING agonist that not only induces innate antiviral immunity against a broad spectrum of viruses but may also stimulate the activation of adaptive immune responses, which is important for the treatment of chronic viral infections and tumors.

New perfluorinated ionomer with improved oxygen permeability for application in cathode polymeric electrolyte membrane fuel cell

Proton Exchange Membrane Fuel Cells (PEM-FC) are considered an alternative to the internal combustion engine for transportation. Despite the existence of relevant demonstrations and few commercial fleets, there are still a series of issues such as the durability of the materials and the cost of platinum which limit a wider application in this field. To this regard, better utilization and consequently loading reduction of Pt in electrodes is of outstanding importance. Herein we report the synthesis, characterization and application of an ionomer in the cathode catalyst layer; thanks to its higher oxygen permeability the quantity of Pt may be minimized. This ionomer is a modification of commercial Aquivion PFSA obtained by incorporation of a third monomer (2,2,4-trifluoro-5-trifluoromethoxy-1,3-dioxole, MDO). Results on cast membrane demonstrate a 20% higher oxygen permeability of the new ionomer compared to Aquivion PFSA E87-05S and Nafion NR212 at 100% RH. Furthermore, an improvement of the performance of a PEM-FC in automotive conditions (RH = 40-70%) is obtained with the assembly where this ionomer is used as cathode binder. In particular polarization curve at 70% RH shows improvement of 20% of power density peak and electrochemical impedance confirms higher oxygen permeability due to lower mass transport resistance than Aquivion.

Water ion adsorption dominates charging at nonpolar polymer surfaces in multivalent electrolytes

Fluoropolymer films, such as Teflon(TM)-TFE [poly(tetrafluoroethylene)], Teflon(TM)-FEP [copolymer of tetrafluoroethylene and hexafluoropropylene], Teflon(TM)-PFA [copolymer of tetrafluoroethylene and perfluoro(propyl vinyl ether)], and PCTFE [poly(chlorotrifluoroethylene)], are reduced by the mild reducing agent benzoin dianion/DMSO, while Teflon(TM)-AF [copolymer of tetrafluoroethylene and perfluoro-2,2-dimethyl-1,3-dioxole] is unreactive. The reduction makes the films adherable toward epoxy resins, the adhesive strength decreasing in the order PCTFE > PFA greater than or equal to FEP > PTFE. Surprisingly, Teflon(TM)-AF films are totally inert except when the TFE content is high, and in these cases, the adhesive strength of Teflon(TM)-AF is close to that of PTFE. The surfaces of PTFE, FEP and PFA films are further modified by first treating these films with the benzoin dianion/DMSO reagent and then with an excess of sodium salts of mercaptans. Reactive PTFE films that are light colored with a metallic luster are formed rather than the dark, metallic color typical of PTFE surface reduction without mercaptan treatment. These films have poor adhesion toward epoxy resins but good adhesion toward gold applied by sputtering. FEP and PFA films behaved similarly. These results are attributed to the incorporation of sulfur onto the reduced surface. Unreduced fluoropolymer films reacted with only sodium mercaptan fail to show C-S bond incorporation. This newly developed method can be used to selectively metallize the fluoropolymer film surface with gold layers and affords very high conductivity of the metallized regions. Quantitatively controllable fluid flows with ballpoint-pen-printed patterns for programmable photo-paper-based microfluidic devices

Regulating the fluid flow in microfluidic devices enables a wide range of assay protocols for analytical applications. A programmable, photo-paper-based microfluidic device fabricated by using a method of cutting and laminating, followed by printing, is reported. The flow distance of fluid in the photo-paper-based channel was linearly proportional to time. By printing silver nanoparticle (AgNP) and poly.4,5-difluoro-2,2-bis.trifluoromethyl)-1,3-dioxole-co-tetrafluoroethylene] (PTFE) patterns on the surface of a photo-paper-based channel, we were able to either increase or decrease the fluid flow in the fabricated microfluidic devices, while maintaining the linearity in the flow distance-time relation. In comparison to the speed of fluid flow in a pristine channel, by using hydrophilic AgNP patterns, we were able to increase the speed in the channel by up to 15 times while we were able to slow the speed by a factor of 3 when using hydrophobic PTFE dots. We then further demonstrated a single-step protocol for detecting glucose and a multi-step protocol for detecting methyl paraoxon (MPO) with our methods in photo-paper-based microfluidic devices. This approach can lead to improved fluid handling techniques to achieve a wide range of complex, but programmable, assays without the need for any additional auxiliary devices for automated operation.
