Cloning of the cytochrome P450 enzyme from Perilla frutescens involved in nothoapiole biosynthesis was written by Baba, Mariko;Ito, Michiho. Phenylpropanoid volatile components are found in various plants and are useful in medicines, health foods, and fragrances. While the pharmacol. actions and toxicities of these compounds have been investigated, there are few reports of the cloning of genes that encode biosynthetic enzymes involved in substituent formation at diverse positions and numbers Previously, using the expressed sequence tag (EST) libraries of pure perilla strains that have been maintained for over 30 years for their oil type, we characterized the P 450 enzyme that produces an intermediate for dillapiole by adding a hydroxy group to myristicin. In this study, we selected a P 450 enzyme involved in nothoapiole biosynthesis from the EST library. Heterologous expression of this enzyme in yeast showed that it is a hydroxylase that synthesizes an intermediate to produce nothoapiole from apiole and dillapiole. The enzyme has high amino acid sequence similarity with a previously cloned enzyme and is categorized into the CYP71D subfamily. Furthermore, we investigated the presence or absence of essential oil components and intermediates believed to be involved in nothoapiole biosynthesis by component anal. of perilla essential oil using GC-MS to help elucidate the biosynthetic pathway of nothoapiole. Only a small number of plant species contain nothoapiole as their principal component and thus few studies have reported the biosynthetic genes involved or the drug efficacy and toxicity of nothoapiole. The present study will aid in understanding the biosynthesis of phenylpropanoid volatile compounds, thereby contributing to further research on potentially useful compounds such as nothoapiole. Myristicin [5-allyl-1-methoxy-2,3-(methylenedioxy)benzene] is the major constituent of the seasoning nutmeg oil and powder. Sometimes myristicin is abused via its ingestion at high doses to cause hallucination. In these high doses, myristicin could cause severe adverse health effects, including convulsion, delirium, and palpitation. Hence there is a strong need for a sensitive method for its anal., such as fluorescence determination Myristicin has a very weak fluorescence and also lacks derivatizable groups like the carboxylic, hydroxyl, or amino group in its structure, which makes its fluorescence derivatization challenging. In this research, we developed a fluorescence labeling method for myristicin based on the Mizoroki-Heck coupling reaction of its terminal alkene with a fluorescent aryl iodide derivative, 4-(4,5-diphenyl-1H-imidazol-2-yl)iodobenzene (DIB-I). Then, we developed an HPLC fluorescence detection method for the determination of myristicin utilizing this labeling reaction. The developed method showed a good linear response for myristicin (r = 0.995) in the range of 0.01-10 μmol/L with excellent sensitivity down to the detection limit of 2.9 nmol/L (9.6 fmol/injection). Finally, the developed method could be successfully applied to determine myristicin content in nutmeg powder, oil samples, and human plasma with simple extraction methods and good recoveries ranging from 89.3 to 106%. Synthesis of Trisaccharides by Hetero-Diels-Alder Welding of Two Monosaccharide Units was written by Himanen, Jatta A.;Pihko, Petri M.. And the article was included in European Journal of Organic Chemistry in 2012.SDS of cas: 171086-52-5 This article mentions the following:

A new strategy for the synthesis of di- and trisaccharides based on the de novo synthesis of the linking saccharide unit is presented. In this strategy, functionalized monosaccharide building blocks already incorporating the glycosidic linkages are welded together using a metal-catalyzed hetero-Diels-Alder (HDA) reaction to generate a new monosaccharide unit between them. The highest yields and selectivities in the HDA reaction were obtained by using chiral Schiff base chromium complexes. Disaccharide products were accessible by reaction of Danishefsky’s diene with acetyl- and benzyl-protected galactoside aldehydes. For the synthesis of trisaccharide products, acetyl-protected glucose or galactose-derived dienes were fused with monosaccharide-derived aldehydes using chromium catalysts for the HDA reaction. The desired trisaccharide products were obtained in moderate to good yields with excellent stereoselectivity. AMPA receptor activation potentiated by the AMPA modulator 1-BCP is toxic to cultured rat hippocampal neurons was written by Yamada, Kelvin A.. And the article was included in Neuroscience Letters in 1998.Application In Synthesis of Benzo[d][1,3]dioxol-5-yl(piperidin-1-yl)methanone This article mentions the following:

The benzoylpiperidine 1-(1,3-benzodioxol-5-ylcarbonyl)-piperidine (1-BCP), and related compounds, potentiate α-amino-3-hydroxy-5-methyl-4-isoxazole proprionic acidergic (AMPAergic) synaptic currents in central neurons, and improve performance of rodents and humans on learning and memory tasks. Their physiol. actions are similar but not identical to thiazides, which also enhance AMPAergic synaptic responses and improve performance of rats in water-maze and passive-avoidance tests. Thiazides also dramatically increase AMPA receptor-mediated neuronal death in vitro and in vivo. Here it was evaluated whether 1-BCP potentiated AMPA receptor-mediated excitotoxicity in hippocampal neuron cultures. Glutamate + MK 801 (to block NMDA receptors) + 1 mM 1-BCP produced neuronal death that was reversed by 10 μM 2,3-dihydroxy-6-nitro-7-sulfamoyl-benzo(F)quinoxaline (NBQX), a selective AMPA receptor antagonist. Polycarbonates with pendant functional groups have attracted much attention due to their capability for further chem. modification and post-polymerization This work describes the synthesis of a poly(carbonate-co-ether) with massive pendant acrylate groups from the copolymerization of glycidyl methacrylate (GMA) with carbon dioxide (CO2), using a nanolamellar zinc-cobalt double metal cyanide complex (Zn-Co(III) DMCC) catalyst. The carbonate linkage content (FCO2) of the poly(carbonate-co-ether) could be varied from 42.2 to 68.0% by changing the polymerization conditions. Of importance, 4-methoxyphenol was applied for regulating the copolymerization It could not only act as an inhibitor for completely depressing the self-polymerization of GMA via free radical polymerization of the double bond, but also modulate the mol. weight of the resultant copolymers. The obtained copolymer had two terminal hydroxyl groups, which were confirmed by the electrospray ionization-tandem mass spectrometry (ESI-MS) technique. A new thermoset with high glass transition temperature (Tg: 105 or 120 °C) and massive carbonate units as well as hydroxyl (or carboxylic) groups was prepared by the curing reaction of the GMA-CO2 copolymer with allyl alc. or acrylic acid in the presence of 2,2′-azobisisobutyronitrile (AIBN). Easy α-alkylation of ketones with alcohols through a hydrogen autotransfer process catalyzed by RuCl2(DMSO)4 was written by Martinez, Ricardo;Ramon, Diego J.;Yus, Miguel. And the article was included in Tetrahedron in 2006.Application of 28657-75-2 This article mentions the following:

The electrophilic α-alkylation of ketones with alcs. is accomplished by a hydrogen autotransfer process catalyzed by RuCl2(DMSO)4. The reaction can produce either simple alkylated ketones or α,β-unsaturated ketones just by choosing the appropriate starting ketones (Me ketones or bicyclic methylenic ketones, resp.), as well as quinolines (by using 2-aminobenzyl alc. derivatives) or the corresponding alc. derivatives by the addition of an extra equivalent of the initial alc. In the last case, after the above alkylation process reduction of the carbonyl compound takes place. A mechanistic study seems to indicate that the process goes through the oxidation of the alcs. with ruthenium (after a previous deprotonation) to yield the corresponding aldehyde and a ruthenium hydride intermediate. In turn, the aldehyde suffers a classical aldol reaction with the starting ketone to form the α,β-unsaturated ketone, which finally is reduced through a Michael-type addition by the aforementioned ruthenium hydride intermediate. PEG grafted polymers are a kind of promising materials, and widely applied in polymer electrolytes, lubricants, biotechnol. and biomedical science. While up to now, systematical researches of the relationship between architecture and conformation of PEG grafted polymers are very rare. Here, we efficiently and successfully prepared a series of PEG grafted polymers with different grafting d., and side chain length by combining ring-opening metathesis polymerization (ROMP) and Copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction, and then characterized the conformations of the synthesized bottlebrush polymers by AFM technique. The AFM images showed that all the polymers P(NB-alkyne)400-g-PEG with low grafting d. were spherical morphol., while the polymers P(NB-2alkyne)400-g-PEG and P(NB-4alkyne)394-g-PEG presented cylindrical morphol., which indicated that only grafting d. could determine the shape of the prepared PNB-g-PEG bottlebrush polymers. The stereoselective syntheses of 7,8,9-trideoxypeloruside A I and a monocyclic peloruside A analog lacking the entire tetrahydropyran moiety II are described. The syntheses proceeded through the PMB-ether of an ω-hydroxy β-keto aldehyde as a common intermediate which was elaborated into a pair of diastereomeric 1,3-syn and -anti diols by stereoselective Duthaler-Hafner allylation and subsequent 1,3-syn or anti reduction One of these isomers was further converted into a tetrahydropyran derivative in a high-yielding Prins reaction, to provide the precursor for bicyclic analog I. Downstream steps for both syntheses included the substrate-controlled addition of a vinyl lithium intermediate to an aldehyde, thus connecting the peloruside side chain to C15 (C13) of the macrocyclic core structure in a fully stereoselective fashion. In the case of monocyclic II macrocyclization was based on ring-closing olefin metathesis, while bicyclic I was cyclized through Yamaguchi-type macrolactonization. The macrolactonization step was surprisingly difficult and was accompanied by extensive cyclic dimer formation. Peloruside A analogs I and II inhibited the proliferation of human cancer cell lines in vitro with micromolar and sub-micromolar IC50 values, resp.
