Condensation of phenacyl bromide with amines gave PhCOCH2NHCH2R [R = 4-MeOC6H4, 3,4-(MeO)2C6H3, 3,4-methylenedioxyphenyl] which were condensed with a chloropyrimidinone to give adducts I. Cyclization of I gave substituted guanine oxides II which were debenzylated to give the title compound (III). A series of alkyl and cycloalkyl substituted guanine oxides were prepared by the same methodol. All the substituted guanine oxides were tested for antileukemic activity and none were superior to III.
