NOVEL 5-ARYLCARBAMOYL-2-METHYLISOXAZOLIDIN-3-YL-3-PHOSPHONATES AS NUCLEOTIDE ANALOGUES

A series of 5-substituted 3-phosphonylated isoxazolidines have been obtained via cycloaddition of N-methyl-C-(diethoxyphosphoryl)nitrone with N-heteroaromatic acrylamides. Good trans/cis diastereoselectivities (d.e. 58-76%) of isomeric (3-diethoxyphosphoryl)isoxazolidines were observed. cis- and trans-Isoxazolidine phosphonates were evaluated for their antiviral activity against a broad range of DNA and RNA viruses but were found inactive. Their cytostatic activity toward L1210, CEM, and HeLa cells was also established, and compounds cis-12r and trans-11r having a 2,2-difluorobenzo[d][1,3]dioxole moiety slightly inhibited proliferation of HeLa cells at IC50 values of 186 and 179 mu M, respectively.

Eco-sustainable synthesis and biological evaluation of 2-phenyl 1,3-benzodioxole derivatives as anticancer, DNA binding and antibacterial agents

The current research and development scenario in medicinal chemistry demands small molecules synthesized in a simple, fast and effective way with enhanced activity and fewer side effects than the existing ones. Therefore, one-pot, microwave assisted green and efficient synthesis of a series of derivatives belonging to 2-phenyl 1,3-benzodioxole (1a-14a) and 2-phenyl 1,3-benzo-dioxole-4-ol (1b-14b) class were carried out and subsequently investigated for their anticancer, antibacterial and DNA binding potential. Compound 3c proved to be the most active one among the screened derivatives possessing anticancer and antibacterial potency greater than the standard reference compound (cisplatin and cinoxacin for anticancer and antibacterial activity, respectively). The most active compound in terms of DNA binding capacity was found to be 5b. A rewarding feature of the work is a facile, convenient, eco friendly one step synthesis of compounds demonstrating attenuated activity against cancer and bacterial cell with an inherent potential of binding to DNA. Subsequently, a hit molecule for further anticancer, antibacterial (compound 3c) and DNA binding studies (compound 5b) was also identified. (C) 2014 King Saud University. Novel amorphous perfluorocopolymeric system: Copolymers of perfluoro-2-methylene-1,3-dioxolane derivatives

Perfluorotetrahydro-2-methylene-furo[3,4-d][1,3]dioxole (monomer I) and perfluoro-2-methylene-4-methoxymethyl-1,3-dioxolane (monomer II) are soluble in perfluorinated or partially fluorinated solvents and readily polymerize in solution or in bulk when initiated by a free-radical initiator, perfluorodibenzoyl peroxide. The copolymerization parameters have been determined with in situ F-19 NMR measurements. The copolymerization reactivity ratios are r(I) = 1.80 and r(II) = 0.80 in 1,1,2-trichlorotrifluoroethane at 41 degrees C and r(I) = 0.97 and r(II) = 0.85 for the bulk polymerization. These data show that this copolymerization pair has a good copolymerization tendency and yields nearly ideal random copolymers. The copolymers have only one glass-transition temperature from 101 to 168 degrees C, depending on the copolymer compositions. Melting endotherms have not been observed in their differential scanning calorimetry traces, and this indicates that all the copolymers with different compositions are completely amorphous. These copolymers are thermally stable (the initial decomposition temperatures are higher than 350 degrees C under an N-2 atmosphere) and have low refractive indices and high optical transparency from UV to near-infrared. Copolymer films prepared by casting were flexible and tough. These properties make the copolymers ideal candidates as optical and electrical materials. (c) 2006 Wiley Periodicals, Inc.

Pulse field gradient NMR study of diffusion of pentane in amorphous glassy perfluorodioxole

Pulse field gradient diffusion measurements of pentane in a random copolymer of tetrafluoroethylene (TFE) and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole (PDD) were made as a function of the time allowed for diffusion to occur. The initial change in echo amplitude at low values of q = gamma deltag was used to determine the apparent diffusion constant. The apparent diffusion constant determined in this way decreases to a constant value as time increases. At a time of 12.4 ms the apparent diffusion constant was 2.2 x 10(-7) cm(2) s(-1), and it decreases to 5.87 x 10(-8) cm(2) s(-1) at 1 s. This change in the apparent diffusion constant was interpreted in terms of morphological structure on the micron length scale in this completely amorphous glassy polymer. This polymer has been considered to have high free volume regions which lead to the observed rapid permeation of gases and vapors. These regions are interspersed in lower free volume regions, and the low free volume regions constitute the majority component. In the pulse field gradient NMR experiment, regions allowing for rapid diffusion interspersed with regions allowing only slow diffusion can lead to changes in the apparent diffusion constant as a function of diffusion time. The data observed in this system were compared with two simple models for the topology of the regions allowing for rapid diffusion: restricted diffusion and tortuous diffusion. The observed behavior is qualitatively similar to tortuous diffusion and can be fit with an equation describing this type of diffusion.

Preparation of Hyflon AD60/PVDF composite hollow fiber membranes for vacuum membrane distillation

Hydrophobic membrane is desired to avoid pore wetting phenomenon in membrane distillation (MD). In the present paper, highly hydrophobic Hyflon AD60/PVDF composite hollow fiber membranes were prepared by coating a copolymer (Hyflon AD60) of tetrafluoroethylene (TFE) and 2,2,4-trifluoro-5-trifluoro methoxy-1,3-dioxole (TTD) on poly(vinylidene fluoride) (PVDF) membranes. The effects of Hyflon AD60 concentration, coating time, heat-treatment temperature and heat-treatment time on the structures and performances of Hyflon AD60/PVDF composite hollow fiber membranes in vacuum membrane distillation (VMD) process were investigated. The results show that the hydrophobicity of PVDF hollow fiber membrane was significantly improved by coating Hyflon AD60. The optimized Hyflon AD60 concentration, coating time, heat-treatment temperature and heat-treatment time were 0.1 wt%, 10-20 min, 40-50 degrees C and 9 h, respectively. Employing NaCl solution with concentration of 35 g x L-1, under a vacuum of 0.09 MPa and feed temperature of 70 degrees C, the water flux and salt rejection of the composite hollow fiber membrane in VMD process reached to approximately 10 kg x m(-2) x h(-1) and higher than 99.9 %, respectively, and kept stable, indicating that Hyflon AD60/PVDF composite membrane is an excellent candidate for operation in VMD operation. Thermal conductivity of amorphous Teflon (AF 1600) at high pressure

Cavity size distributions in high free volume glassy polymers by molecular simulation

Two very permeable polymers, poly(1-trimethylsilyl-1-propyne) (PTMSP) and a random copolymer of tetrafluoroethylene and 2.2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole (TFE/BDD), have very similar and large fractional free volumes (FFV), but very different permeabilities. Using atomistic models, cavity size (free volume) distributions determined by a combination of molecular dynamic and Monte Carlo methods are consistent with the observation that PTMSP is more permeable than TFE/BDD. The average spherical cavity size in PTMSP is 11.2 Angstrom whereas it is only 8.2 Angstrom in TFE/BDD. These cavity size distributions determined by simulation are also consistent with free volume distributions determined by positron annihilation lifetime spectroscopy.
