New perfluorinated ionomer with improved oxygen permeability for application in cathode polymeric electrolyte membrane fuel cell

Proton Exchange Membrane Fuel Cells (PEM-FC) are considered an alternative to the internal combustion engine for transportation. Despite the existence of relevant demonstrations and few commercial fleets, there are still a series of issues such as the durability of the materials and the cost of platinum which limit a wider application in this field. To this regard, better utilization and consequently loading reduction of Pt in electrodes is of outstanding importance. Herein we report the synthesis, characterization and application of an ionomer in the cathode catalyst layer; thanks to its higher oxygen permeability the quantity of Pt may be minimized. This ionomer is a modification of commercial Aquivion PFSA obtained by incorporation of a third monomer (2,2,4-trifluoro-5-trifluoromethoxy-1,3-dioxole, MDO). Results on cast membrane demonstrate a 20% higher oxygen permeability of the new ionomer compared to Aquivion PFSA E87-05S and Nafion NR212 at 100% RH. Furthermore, an improvement of the performance of a PEM-FC in automotive conditions (RH = 40-70%) is obtained with the assembly where this ionomer is used as cathode binder. In particular polarization curve at 70% RH shows improvement of 20% of power density peak and electrochemical impedance confirms higher oxygen permeability due to lower mass transport resistance than Aquivion.

Potential Antileptospiral Constituents from Phyllanthus amarus

Background: Phyllanthus amarus (PA) is a well-known herb for its medicinal properties and widely used worldwide. PA has a significant role in Indian Ayurveda system of medicine for treating various ailments such as gonorrhea, menorrhagia, and other genital infections. Objectives: The aim of the study is to investigate antileptospiral activity and isolate the potential antileptospiral constituents of the methanol extract of PA (MPA). Materials and Methods: The primary pharmacological tests for leptospirosis were investigated by test tube dilution technique and microdilution technique. To examine the morphogenesis of experimental leptospirosis by morphologic and histological methods, albino mice were inoculated intraperitoneally with Leptospira interrogans sero group Icterohaemorrhagiae strains. Results: The activity-guided repeated fractionation for MPA through silica gel column chromatography yielded three compounds that exhibited antioxidant and in vitro, in vivo, and in silico antileptospiral activities. Based on diverse physicochemical and spectroscopic analyses (viz., 13C NMR,1H NMR, ultraviolet [UV], IR, and mass spectroscopy), the potential constituents were elucidated as 5-(3-(3,4-dimethoxybenzyl)-4-methoxy-2-(methoxymethyl)butyl)4,7- dimethoxybenzo[d][ 1,3] dioxole(C1), 1-(3-( 3,4- dimethoxybenzyl)- 4-methoxy- 2-(methoxymethyl) butyl)- 2,3,4,5tetramethoxybenzene(C2), and 4-(3-(3,4dimethoxybenzyl)-4-methoxy-2-(methoxymethyl)butyl)-3,6-dimethoxybenzene-1,2-diol (C3). The histopathological examinations of both kidney and liver showed promising activity with C3 at 75 and 100 mu g/mL, respectively. Conclusion: The in vitro, in vivo, and in silico studies revealed that benzo methoxy class of compounds has great potential as antileptospiral agents.

The synthesis and application of (E)-N ‘-(benzo[d]dioxol-5-ylmethylene)-4-methyl-benzenesulfonohydrazide for the detection of carcinogenic lead

In this study, noble ligands of (E)-N ‘-(benzo[d]dioxol-5-ylmethylene)-4-methyl-benzenesulfonohydrazide (BDMMBSH) were prepared via a simple condensation method using benzo-[d][1,3]-dioxole carbaldehyde, benzenesulfonylhydrazine (BSH), and 4-methyl-benzenesulphonylhydrazine (4-MBSH) in good yield, which were crystallized in acetone, EtOAc, and EtOH. The BDMMBSH derivatives were characterized using different spectroscopic techniques, such as H-1-NMR, C-13-NMR, FTIR, and UV-Vis spectroscopy, and their crystal structures were analyzed using the single crystal X-ray diffraction method (SCXRDM). Subsequently, the BDMMBSH compounds were used for the significant detection of the carcinogenic heavy metal ion, lead (Pb2+), via a reliable electrochemical approach. A sensitive and selective Pb2+ sensor was developed via the deposition of a thin layer of BDMMBSH on a GCE with the conducting polymer matrix Nafion (NF). The sensitivity, LOQ, and LOD of the proposed sensor towards Pb2+ were calculated from the calibration curves to be 2220.0 pA mu M-1 cm(-2), 320.0 mM, and 96.0 pM, respectively. The validation of the BDMMBSH/GCE/NF sensor probe was performed via the selective determination of Pb2+ in spiked natural samples with a satisfactory and rational outcome.

Molecular design of triarylamine dyes incorporating phenylene spacer and the influence of alkoxy substituent on the performance of dye-sensitized solar cells

The effects of alkoxy substituent upon the photophysical, electro-chemical characteristics and performance of dye-sensitized solar cells are investigated. For a typical device, the MX13-based cell affords an overall power conversion efficiency (eta) of 7.02%, with short-circuit photocurrent density (J(SC)), open-circuit voltage (V(OC)) and fill factor (if) of 15.5 mA cm(-2), 697 mV and 0.65, respectively. Metabolite profiling of C-14-omacetaxine mepesuccinate in plasma and excreta of cancer patients

1. Omacetaxine mepesuccinate (hereafter referred to as omacetaxine) is a protein translation inhibitor approved by the US Food and Drug Administration for adult patients with chronic myeloid leukemia with resistance and/or intolerance to two or more tyrosine kinase inhibitors. 2. The objective was to investigate the metabolite profile of omacetaxine in plasma, urine and faeces samples collected up to 72h after a single 1.25-mg/m(2) subcutaneous dose of C-14-omacetaxine in cancer patients. 3. High-performance liquid chromatography mass spectrometry (MS) (high resolution) in combination with off-line radioactivity detection was used for metabolite identification. 4. In total, six metabolites of omacetaxine were detected. The reactions represented were mepesuccinate ester hydrolysis, methyl ester hydrolysis, pyrocatechol conversion from the 1,3-dioxole ring. Unchanged omacetaxine was the most prominent omacetaxine-related compound in plasma. In urine, unchanged omacetaxine was also dominant, together with 4′-DMHHT. In feces very little unchanged omacetaxine was found and the pyrocatechol metabolite of omacetaxine, M534 and 4′-desmethyl homoharringtonine (4′-DMHHT) was the most abundant metabolites. 5. Omacetaxine was extensively metabolized, with subsequent renal and hepatic elimination of the metabolites. The low levels of the metabolites found in plasma indicate that the metabolites are unlikely to contribute materially to the efficacy and/or toxicity of omacetaxine.

Collagen modified with epoxidized safrole for improving antibacterial activity

An epoxidized safrole, 5-(oxiran-2-ylmethyl)-benzo[d][1,3] dioxole (OYBD), was synthesized and employed to modify collagen for improving its antibacterial activity. The interaction between collagen and OYBD, and the structure/properties of the modified collagen were investigated in detail. The results indicated that the OYBD-modified collagen showed a higher de-nature temperature (Td, 90.2 degrees C), improved hydrophobic properties (contact angle from 84.2 degrees to 89.1 degrees) and enhanced tensile strength (6.2-11.0%) without destroying its triple helix structure. From observation of scanning electron microscopy (SEM), a higher density of intertwining morphology and a more stable network structure were observed, which was consistent with improved tensile strength and reduced breaking extension stress. The antibacterial test and LIVE/DEAD Baclight bacterial viability assay illustrated that the modified collagen exhibited excellent antibacterial activity to both Gram-negative and Gram-positive bacteria. Furthermore, the OYBD-modified collagen still exhibited cytocompatibility, supporting human fibroblast proliferation, which holds a great potential for developing antibacterial collagen-based biomaterials.

Discovery of natural alkaloids as potent and selective inhibitors against human carboxylesterase 2

Human Carboxylesterase 2A (hCES2A), one of the most important serine hydrolases, plays crucial roles in the hydrolysis and the metabolic activation of a wide range of esters and amides. Increasing evidence has indicated that potent inhibition on intestinal hCES2A may reduce the excessive accumulation of SN-38 (the hydrolytic metabolite of irinotecan with potent cytotoxicity) in the intestinal tract and thereby alleviate the intestinal toxicity triggered by irinotecan. In this study, more than sixty natural alkaloids have been collected and their inhibitory effects against hCES2A are assayed using a fluorescence-based biochemical assay. Following preliminary screening, seventeen alkaloids are found with strong to moderate hCES2A inhibition activity. Primary structure-activity relationships (SAR) analysis of natural isoquinoline alkaloids reveal that the benzo-1,3-dioxole group and the aromatic pyridine structure are beneficial for hCES2A inhibition. Further investigations demonstrate that a steroidal alkaloid reserpine exhibits strong hCES2A inhibition activity (IC50 = 0.94 mu M) and high selectivity over other human serine hydrolases including hCES1A, dipeptidyl peptidase IV (DPP-IV), butyrylcholinesterase (BChE) and thrombin. Inhibition kinetic analyses demonstrated that reserpine acts as a noncompetitive inhibitor against hCES2A-mediated FD hydrolysis. Molecular docking simulations demonstrated that the potent inhibition of hCES2A by reserpine could partially be attributed to its strong sigma-pi and S-pi interactions between reserpine and hCES2A. Collectively, our findings suggest that reserpine is a potent and highly selective inhibitor of hCES2A, which can be served as a promising lead compound for the development of more efficacious and selective alkaloids-type hCES2A inhibitors for biomedical applications.

Inhalation toxicity of Dioxole and Dioxolane compounds in the rat

Four chemicals (Dioxole 418, Dioxolane 418, Dioxolane 416 and Dioxolane 456) which are used as stabilizers in highresolution image were tested or both their acute and repeated inhalation toxicity in the rat using nose-only exposures. Acute studies determined the lethal concentrations following a single 4-hour exposure, repeated exposure inhalation studies determined the potency and target tissue(s) following 6-hour/day exposures, 5 days/week for 2 weeks. Each of the chemicals was at least mildly toxic acutely with approximate lethal concentrations of > 1,500 ppm for Dioxole 418, 1,300 ppm for Dioxolane 418, 1,700 ppm for Dioxolane 416, and 4,300 ppm for Dioxolane 456. No specific unusual clinical signs of response were seen in the rats exposed acutely. Repeated exposures with Dioxole 418 and Dioxolane 418 resulted in no evidence of toxicity with NOAEL’s being 440 and 500 ppm respectively (the highest concentrations tested). Repeated exposures to 250 ppm Dioxolane 456 were not tolerated with mortalities observed after exposure. Severe bone marrow hypoplasia along with reductions in platelet and neutrophil counts were observed at this concentration with less severe hemopoietic changes seen also at 10 and 51 ppm. The no-effect level for Dioxolane 456 was determined to be 10 ppm in female rats and I ppm in males. The same hemopoietic effects were seen with Dioxolane 416 at exposures of 53 ppm or greater in males but not in females exposed to 53 ppm Dioxolane 416. Hepatocellular hypertrophy and depression of serum alkaline phosphatase activity were seen in male rats exposed to 500 but not 53 ppm Dioxolane 416. Testicular degeneration was also seen in rats exposed to 500 ppm Dioxolane 416. The NOAEL was 5 ppm for the chemical.
