In this work, a high molecular weight copolymer of tetrafluoroethylene and perfluoro-4-trifluoromethoxy-1,3-dioxole (HYFLON (R) AD60) and two perfluoropolyethers (PFPEs) containing ammonium phosphate or triethoxysilane functionalities, FLUOROLINK (R) F10 and FLUOROLINK (R) S10 respectively, have been evaluated as protective coatings that can be easily applied on anodized titanium surfaces. Water and n-dodecane contact angle measurements have been recorded by using the sessile drop method for bare and coated surfaces in order to determine the hydrophobic and oleophobic properties of the coatings. The UV-stability of coatings have been studied by Fourier transform infrared spectroscopy (FT-IR) analyses and by observing the variation of water contact angles on coated substrates before and after UV irradiation at regular time intervals. The thickness of the fluorinated films has been measured by ellipsometry and by weight evaluation. Preliminary tests of the adhesion between films and substrate have been conducted. The permeability of glassy polymers to ozone was measured for the first time. The measurements were made on amorphous AF Teflons (DuPont), the copolymers of bis(trifluoromethyl-4,5-difluoro-1,3-dioxole) and tetrafluoroethylene with a cyclic comonomer content of 87 (AF2400) and 65 (AF1600) mol %, at 20-60 degrees C and an ozone partial pressure of 0-5.5 kPa in an ozone-oxygen mixture. It was shown that the obtained permeability coefficients depended neither on the amount of ozone in the mixture nor on the polymer film thickness (5-15 mu m) and were equal to 2160 and 525 Barrer (at 20 degrees C) for Teflon AF2400 and AF1600, respectively. The temperature dependences of permeability coefficients are qualitatively different for individual copolymers. The permeability of Teflon AF2400 decreased with increasing temperature (the apparent activation energy of permeability is negative, -9.3 kJ/mol), whereas normal behavior was observed for Teflon AF1600, i.e., the permeability coefficient grew with temperature. (The activation energy of permeability is 6.4-7.0 kJ/mol for films of various thickness.) This difference is due to an abnormally low activation energy of diffusion in AF2400, which has a high free volume.

Pruinosanone A (1), a novel spirochromone, was isolated from the roots of Caragana pruinosa. Two biogenetically related isoflavone intermediates, pruinosanones B and C (2 and 3), were also isolated, together with five known analogs identified as 3-hydroxy-9-methoxypterocarpan (4), 7,2′-dihydroxy-4′-methoxyisoflavanol (5), retusin-8-methylether (6), 7,2′-dihydroxy-8,4′-dimethoxy isoflavone (7) and 7,3′-dihydroxy-8,4′-dimethoxy isoflavone (8). The structures of 1-3 were elucidated based on extensive spectroscopic methods. Notably, 1 is the first example of a spirochromone possessing an unprecedented pentacyclic skeleton containing a spiro[benzo[d][1,3] dioxole-2,3′-chroman]-4′-one motif, which was confirmed by X-ray diffraction analysis. A plausible biosynthetic pathway for 1 was also proposed. Compounds 1-8 were tested for their ability to inhibit nitric oxide (NO) production in LPS-induced RAW 264.7 macrophages, and compounds 1-3 were the most potent inhibitors of NO production, with IC50 values of 1.96, 1.93 and 1.58 mu M, respectively. A structure-activity relationship analysis revealed that the fused 2-isopropenyl-2,3-dihydrofuran moiety plays a vital role in the potency of these compounds. Moreover, 1 was found to significantly inhibit inducible nitric oxide synthase (iNOS) protein expression, which accounts for the potent inhibition of NO production by this spirochromone.

Electrochemical Behavior and Antioxidant Activity of Hibalactone

Hydrocotyle umbellata L. is a groundcover plant that has been used in the folk medicine of Argentina, Brazil, Cuba e India, mainly to reduce inflammatory processes. The lignan, hibalactone, can be isolated from the roots of the plant. In the anti-inflammatory and antioxidant activity of such natural products, it is believed that the mechanism of action involves the butyrolactone ring, which would act as free pharmacophore. In this study, electrochemical, spectrometric and computational chemistry approaches were used to evaluate the lignan antioxidant activity and electro-oxidation pathway. Electrochemical measurements were performed in a 3 mL cell, operated in a three electrode system, consisting in an Ag/AgCl reference electrode, platinum as counter electrode and glassy carbon electrode was used as working electrode. Radical scavenging assays were performed with DPPH and ABTS. In the computational chemistry approach geometry, optimization was performed with semi empirical calculations (PM3) and the orbitals/charges calculations were performed by the extended Huckel method. The voltammetry showed a very slight variation with pH alteration, which can be justified by lactone hydrolysis that occurs in acidic and basic pH. In the HOMO and total charge density distribution it was possible to observe that the higher prevalence was in the benzo[1,3]dioxole rings and in the alkenes/alkanes in proximity with the lactone ring. The hibalactone displayed a single quasi-reversible electro-oxidation. Given the HOMO and charge density distribution in the lignan, more so the single oxidation in the evaluated potential, it is reasonable to infer that the alkene bonded with the lactone ring was a probable site of oxidation.

Antiproliferative activity of the Antrodia camphorata secondary metabolite 4,7-dimethoxy-5-methylbenzo[d][1,3]dioxole and analogues (Reprinted from Fitoterapia, vol 123, pg 9-12, 2017)

Both the traditional Chinese medicinal fungus, Antrodia camphorata, and its secondary metabolite, 4,7-dimethoxy-5-methylbenzo[d][1,3]dioxole, have been reported to possess promising anticancer activity. In this work the natural product and analogues bearing more polar substituents were synthesised and assessed for antiproliferative activity in the NCI-60 screen. Although each compound inhibited the growth of some cell lines at 10 mu M, none had sufficient activity to warrant further investigation.

Selective, competitive and mechanism-based inhibitors of human cytochrome P450 2J2

Twenty five derivatives of the drugs terfenadine and ebastine have been designed, synthesized and evaluated as inhibitors of recombinant human CYP2J2. Compound 14, which has an imidazole substituent, is a good non-competitive inhibitor of CYP2.12 (IC50 – 400 nM). It is not selective towards CYP2.12 as it also efficiently inhibits the other main vascular CYPs, such as CYP2B6, 2C8. 2C9 and 3A4: however, it could be an interesting tool to inhibit all these vascular CYPs. Compounds 4, 5 and 13, which have a propyl, allyl and benzo-1,3-dioxole terminal group, respectively, are selective CYP2J2 inhibitors. Compound 4 is a high-affinity, competitive inhibitor and alternative substrate of CYP2J2 (K-i = 160 50 nM). Compounds 5 and 13 are efficient mechanism-based inhibitors of CYP2J2 (k(inact)/K-i values similar to 3000 L mol(-1) s(-1)). Inactivation of CYP2J2 by 13 is due to the formation of a stable iron-carbene bond which occurs upon CYP2J2-catalyzed oxidation of 13 with a partition ratio of 18 +/- 3. These new selective inhibitors should be interesting tools to study the biological roles of CYP2J2. Published by Elsevier Inc.

Degradation mechanism and materials for 157 nm pellicles

A great deal of resources has been invested by the semiconductor industry as a whole to make ready 157 nm as the next lithography technology node. Despite of all this effort serious infrastructure issues remain to be solved. Perhaps the first one is the availability of CaF2, but a close second is a suitable soft-pellicle material. It has been previously reported(1-4) that standard 193 nm materials, like Teflon AF(R) and Cytop(R), fail catastrophically upon exposure at 157 nm radiation; and that their transmission is very poor to non-existent. In this paper we report data showing that these materials have higher transmission and lifetimes than previously thought 2. The physical lifetime of Cytop(R) of nearly 500 J/cm(2) is remarkably high, although the transmission varies as a function dose suggesting physical and chemical changes from the onset. For Teflon AF(R) the transmission is a more complex function of exposure dose, suggesting competing mechanisms. From experiments run with PVDF (polyvinyl difluoride), we conclude that the widely reported photodarkening effect is due to the presence of hydrogen in close proximity to a fluorine atom. From IR spectra we conclude that the dioxole moeity in Teflon AF(R) undergoes a series of photochemical reactions that lead to the physical destruction of the polymer.

Identification, Separation and Characterization of Process-Related Impurities of Bifendate Derivative (DB-6), an Investigational Agent Combating Acute Liver Failure

DB-6, with an IUPAC name of (Z)-5-(2,4-di-tert-butyl-6-((2,4-dioxothiazolidin-5-ylidene)methyl)phenyl)-5′-methyl-7,7′-dimethoxy-[4,4′-bibenzo[ d][1,3] dioxole]-5,5′-dicarboxylate,is an investigational agent for acute liver failure. The focus of this study is the identification and characterization of major unknown impurities in DB-6 bulk drug samples. Four major impurities of DB-6 were detected by a high-performance liquid chromatography (HPLC) method and designated as IMP-I, IMP-II, IMP-III and IMP-IV. Accurate masses of these impurities were determined by using a Q-TOF mass spectrometer. Based on chromatographic, spectrometric data and plausible chemical transformation mechanism, the structures of IMP-I, IMP-II, IMP-III and IMP-IV were identified, respectively, as bifendate, (E)-5-(2,4-ditert- butyl-6-((2,4-dioxothiazolidin-5-ylidene) methyl) phenyl)-5′-methyl-7,7′- dimethoxy-[4,4′-bibenzo[d][1,3] dioxole]-5,5′-dicarboxylate, (Z)-5( 3,5-di-tert-butyl-2-hydroxybenzylidene) thiazolidine-2,4-dione and bis(2,4-di-tert-butyl-6-((Z)-(2,4-dioxothiazolidin-5-ylidene) methyl)phenyl)- 7,7′-dimethoxy-[4,4′-bibenzo[d][1,3] dioxole]-5,5′-icarboxylate. The impurities were isolated by preparative-HPLC or preparative high-speed counter-current chromatography and their structures were confirmed by NMR spectroscopy.
