Temperature dependence of the free volume in amorphous teflon AF1600 and AF2400: A pressure-volume-temperature and positron lifetime study

The microstructure of the free volume and its temperature dependence were studied for amorphous Teflon AFs using pressure-volume-temperature experiments (PVT) and positron annihilation lifetime spectroscopy (PALS). Two copolymers of tetrafluoroethylene and 2,2-bis(trifluoromethyl)-4,5-difluoro-1,3-dioxole units with a composition of 35:65 (AF 1600, T-g = 160 degrees C) and 13:87 mol parts (AF 2400, Tg = 240 degrees C) were investigated after drying the as-received materials. The results are compared with those for CYTOP (T-g = 105 degrees C) and the perfluoroelastomer PFE (T-g = -2 degrees C) as well as for conventional (non-fluorinated) polymers. The PVT data were fitted by the Tait equation and the Simha-Somcynsky (S-S) equation of state (eos). From the latter one the hole fraction h and the specific hole free volume, V-f = hV, were determined. From the PALS data published recently by some of the authors (Rudel et a]. Macromolecules 2008, 41, 788) the free volume hole size distribution characterized by its mean, , and standard deviation, sigma(h), was calculated. The identification of V-f = N-h’ from PALS with V-f = hV from S-S eos opens the way to estimate the specific hole density N-h’. We give evidence for the exceptional large local (, sigma(h)) and fractional (h) hole free volume in the AF polymers already in the glassy state and relate this to the properties of the particular structure of the heterocyclic dioxole comonomer containing two exocyclic -CF3 groups and to the high dioxole fraction in these copolymers. Correlations between the volume parameters at T-g (, sigma(hg), and h(g)) and the value of T-g or the mass of the S-S mer, respectively, were analyzed. AF2400 shows in some aspects a particularly irregular behavior. Possible limits for the application of the S-S theory to our polymers and complications in interpretation of the PALS data are discussed.

LOW-FREQUENCY VIBRATIONAL-SPECTRA AND RING-PUCKERING POTENTIAL-ENERGY FUNCTION OF 1,3-DIOXOLE – A CONVINCING DEMONSTRATION OF THE ANOMERIC EFFECT

The gas-phase far-infrared spectrum shows a series of eleven single-quantum-jump and three triple-quantum-jump transitions in the 40-330 cm-1 region. The low-frequency Raman spectrum exhibits eight ring-puckering transitions corresponding to DELTAupsilon = 2 or 4 transitions in the 160-300-cm-1 region. The ring-puckering potential energy function was determined to be V(cm-1) = (1.59 x 10(6))x4 – (4.18 x 10(4))x2, where x is the ring puckering coordinate in angstroms. This function indicates that the molecule is puckered with a barrier to planarity of 275 cm-1 and a bending angle of 24-degrees. The unexpected nonplanarity of 1,3-dioxole is attributed to the anomeric effect which can be present in molecules with O-C-O linkages. Molecular mechanics calculations utilizing the MM3 parametrization predict a planar structure for this molecule. However, the anomeric effect dictates that each of the =C-O-C-O torsional angles should have a strong desire to increase from 0-degrees toward 90-degrees in order to optimize n-sigma* overlap. When the MM3 force field is modified to reflect this by increasing the magnitude of the 2-fold torsional potential energy term V2 to -5.965 kcal/mol, a reasonably good agreement between the experimental and molecular mechanics potential functions can be obtained.

Block copolymer compatibilized polymer: fullerene blend morphology and properties

Recent studies have shown the role of block copolymer as compatibilizer in tuning the phase separated morphology of the active layer so as to improve the overall photovoltaic efficiency of organic photovoltaic (OPV) devices. Here, we substantiate this observation by investigating the role of a rod-coil block copolymer poly-(3-hexylthiophene)-b-polystyrene (P3HT-b-PS) as compatibilizer in influencing the blend morphology and device performance of several polymer:fullerene blend systems. Fullerene derivatives N-(3-methoxypropyl)-2-(carboxyethyl)-5-(4-cyanophenyl) fulleropyrrolidine (NCPF) and N-(3-methoxypropyl)-2-(carboxyethyl)-5-(5, 5-difluorobenzo-dioxole) fulleropyrrolidine (FFNCPF) were synthesized using Prato reaction, while P3HT-b-PS copolymer was synthesized using the combination of Grignard metathesis, ATRP, and click chemistry. The addition of P3HT-b-PS in P3HT:PCBM blend led to the formation of more homogenous structure compared to the pure P3HT:PCBM blend. Also, a reduction in domain size was observed in P3HT: FFNCPF system upon P3HT-b-PS addition which can be attributed to the compatibilization effect of BCP. Incorporation of P3HT-b-PS block copolymer was found to effectively alter the thin film nanostructure of polymer/fullerene derivative blends and polymer crystalline structure. Maximum enhancement in power conversion efficiency (PCE) of compatabilized P3HT:PCBM blend system was noticed followed by P3HT:FFNCPF and P3HT:NCPF. The moderate improvement in photovoltaic properties can be correlated with face-on orientation of P3HT crystallites and the segregation of the fullerene domains at the bulk heterojunction (BHJ) cathode interface which facilitates the efficient charge collection as respective electrodes. The experimental data analyzed with a Gaussian barrier produce a barrier of 49 cm(-1) as compared to a value of 47 cm(-1) computed using the CCSD/6-311++G(d,p) basis set. The experimental value for the deuteride was determined to be 41 cm(-1) from the one-dimensional approximation. All MP2 and DFT computations for the 1,1-difluoro derivative predict a planar structure whereas the MP2 computation when used with triple-zeta basis set predicts a barrier of 13 cm(-1) for the chloride. Vasodilator and antihypertensive effects of a novel N-acylhydrazone derivative mediated by the inhibition of L-type Ca2+ channels

New bioactive N-acylhydrazone derivatives synthesized from safrole previously have been found to promote intense vasodilation and antihypertensive activity. In this study, we describe the synthesis and the cardiovascular effects of the new N-acylhydrazone derivative (E)-N-methyl-N ‘-(thiophen-3-ylmethylene)benzo[d][1,3]dioxole-5-carbohydrazide (LASSBio-1289). Thoracic aorta and left papillary muscles from Wistar-Kyoto (WKY) rats and spontaneously hypertensive rats (SHR) were prepared for isometric tension recording. LASSBio-1289 promoted relaxation of endothelium-intact and denuded aortic rings with respective pIC(50) (-log IC50) values of 5.07 +/- 0.09 and 4.26 +/- 0.09 (P<0.001) for WKY rats and 5.43 +/- 0.05 and 5.58 +/- 0.07 (P>0.05) for SHR. The vasodilator activity of LASSBio-1289 was increased in the KCl-contracted aorta. LASSBio-1289 attenuated the contracture elicited by Ca2+ in depolarized aorta from both WKY rats and SHR. In endothelium-intact aorta from WKY rats, LASSBio-1289-induced relaxation was unchanged after incubation with propranolol, ZM 241385, atropine, diphenhydramine, and HOE140, but was significantly reduced by L-NAME and ODQ. LASSBio-1289 decreased papillary muscles contractility only at concentrations above 200 mu m. Acute intravenous injection of LASSBio-1289 (3mg/kg) produced a significant hypotensive response in SHR but not in WKY rats, suggesting its antihypertensive profile. The antihypertensive effect was also observed in SHR during 14days of intraperitoneal and oral administration. In conclusion, our data demonstrated that LASSBio-1289 induces both endothelium-independent vasorelaxation involving the inhibition of Ca2+ influx through L-type Ca2+ channels in aorta from WKY rats and SHR, and endothelium-dependent relaxation mediated by the NO/cyclic GMP pathway in WKY rats.

Experimental and Computational Evaluation of Piperonylic Acid Derived Hydrazones Bearing Isatin Moieties as Dual Inhibitors of Cholinesterases and Monoamine Oxidases

A set of piperonylic acid derived hydrazones with variable isatin moieties was synthesized and evaluated for their inhibitory activity against the enzymes acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and monoamine oxidases A and B (MAO-A/B). The results of in vitro studies revealed IC50 values in the micromolar range, with the majority of the compounds showing selectivity for the MAO-B isoform. N-[2-Oxo-1-(prop-2-ynyl)indolin-3-ylidene]benzo[d][1,3]dioxole-5-carbohydrazide (3) was identified as a lead AChE inhibitor with IC50=0.052 +/- 0.006 mu m. N-[(3E)-5-chloro-2-oxo-2,3-dihydro-1H-indol-3-ylidene]-2H-1,3-benzodioxole-5-carbohydrazide (2) was the lead MAO-B inhibitor with IC50=0.034 +/- 0.007 mu m, and showed 50 times greater selectivity for MAO-B over MAO-A. The kinetic studies revealed that compounds 2 and 3 displayed competitive and reversible inhibition of AChE and MAO-B, respectively. The molecular docking studies revealed the significance of hydrophobic interactions in the active site pocket of the enzymes under investigation. Further optimization studies might lead to the development of potential neurotherapeutic agents.

Interplay of structure and reactivity in a most unusual furan Diels-Alder reaction

Difluorinated alkenoate ethyl 3,3-difluoro-2-(N, N-diethylcarbamoyloxy)-2-propenoate reacts rapidly and in high yield with furan and a range of substituted furans in the presence of a tin(IV) catalyst. Non-fluorinated congener 2-(N, N-diethylcarbamoyloxy)-2-propenoate fails to react at all under the same conditions. They reveal a highly polar transition state, which is stabilized by the Lewis acid catalyst SnCl4 and by polar solvents. In the presence of both catalyst and solvent, a two-step reaction is predicted, corresponding to the stepwise formation of the two new carbon-carbon bonds via transition states which have similar energies in all cases. Our experimental observations of the lack of reaction of the non-fluorinated dienophile, the stereochemical outcomes, and the rate acceleration accompanying furan methylation are all well predicted by our calculations. The calculated free energy barriers generally correlate well with measured reaction rates, supporting a reaction mechanism in which zwitterionic character is developed strongly. An in situ ring opening reaction of exo-cycloadduct ethyl exo-2-(N, N-diethylcarbamoyloxy)-3,3-difluoro-7-oxabicyclo[2.2.1] hept-5-enyl-2-endo-carboxylate, which results in the formation of cyclic carbonate ethyl 4,4-difluoro-5-hydroxy-2-oxo-5,7a-dihydro-4H-benzo[1,3] dioxole-3a-carboxylate by a Curtin-Hammett mechanism, has also been examined. Substantial steric opposition to Lewis acid binding prevents carbonate formation from 2-substituted furans.

SYNTHESIS AND PHARMACOLOGICAL EXAMINATION OF BENZOFURAN, INDAN, AND TETRALIN ANALOGS OF 3,4-(METHYLENEDIOXY)AMPHETAMINE

Benzofuran, indan and tetrahydrohaphthalene analogs of 3,4-(methylenedioxy)amphetamine (MDA) were prepared in order to examine the role of the dioxole ring oxygen atoms of MDA in interacting with the serotonin and catecholamine uptake carriers. The series of compounds was evaluated for discriminative stimulus effects in rats trained to discriminate saline from the training drugs (S)-(+)-MBDB (1c), MMAI (3), and (S)-(+)-amphetamine and for the ability to inhibit the uptake of [H-3]serotonin, [H-3]dopamine, and [H-3]norepinephrine into crude synaptosome preparations. Behaviorally, the benzofuran and indan analogs 4-6 produced similar discriminative cues, whereas the tetralin derivative 7 did not fully substitute for the training drugs. The results in the in vitro pharmacology studies indicate that selectivity for 5-HT versus catecholamine uptake carriers may be modulated by the position and orientation of ring oxygen atoms. However, the nonoxygenated isostere 6 possessed high potency at all uptake sites examined. Enlargement of the saturated ring by one methylene unit to give the tetralin derivative resulted in a large (3-4-fold) reduction in activity at catecholamine sites.

In this paper is presented a study on the effects of physical treatments, namely electron beam irradiation or fluorination, on a perfluoropolymer copolymer of tetrafluoroethylene with 2,2,4-trifluoro-5-trifluoromethoxy-1,3-dioxole (Hyflon (R) AD copolymer). The analysis has been carried out by means of IR spectroscopy and quantum chemical modeling based on density functional theory; this combined experimental/ theoretical approach has proven effective for the interpretation of previously unassigned IR bands, which are associated to functional groups generated by polymer degradation and chain scission. We performed a systematic screening of chemical groups and structures compatible with degradation pathways that are possible from the chemical point of view: the chemical mechanisms and the correlation with the spectroscopic experimental data (both frequency and intensity) provide guidelines in understanding the phenomena. Moreover, the spectroscopic experimental/theoretical and chemical approaches allowed us to identify some chemical structures responsible for the unassigned IR bands in the C=O stretching frequency region above 1800 cm(-1), which is typical for carbonyl groups in fluorinated systems.
