The vasodilator activity of LASSBio-1289 was increased in the KCl-contracted aorta. LASSBio-1289 attenuated the contracture elicited by Ca2+ in depolarized aorta from both WKY rats and SHR. In endothelium-intact aorta from WKY rats, LASSBio-1289-induced relaxation was unchanged after incubation with propranolol, ZM 241385, atropine, diphenhydramine, and HOE140, but was significantly reduced by L-NAME and ODQ. LASSBio-1289 decreased papillary muscles contractility only at concentrations above 200 mu m. Acute intravenous injection of LASSBio-1289 (3mg/kg) produced a significant hypotensive response in SHR but not in WKY rats, suggesting its antihypertensive profile. The antihypertensive effect was also observed in SHR during 14days of intraperitoneal and oral administration. In conclusion, our data demonstrated that LASSBio-1289 induces both endothelium-independent vasorelaxation involving the inhibition of Ca2+ influx through L-type Ca2+ channels in aorta from WKY rats and SHR, and endothelium-dependent relaxation mediated by the NO/cyclic GMP pathway in WKY rats.

Background: Thiourea derivatives bearing sulfonamide moiety are well known for their anticancer activity. We also find that even with these design principles, it is advantageous to avoid small rings, as arise in the cyclobutanes. These results suggest a steric component to transparency in addition to the importance of alternation. Upon irradiation these polymers undergo photochemical darkening and therefore none has demonstrated the kilojoule radiation durability lifetimes required to be commercially attractive. This is likely because these exposure lifetimes require every bond to absorb similar to10 photons, each photon having an energy roughly twice common bond energies. We have studied intrinsic (composition, molecular weight) and extrinsic (trace metals, impurities, environmental contaminants, oxygen, water) contributions to optical absorption and photochemical darkening in these polymers. Studies of photochemical darkening in model molecules illustrate the dynamics of photochemical darkening and that appreciable lifetimes can be achieved in fluorocarbons. To a first approximation the polymers that have lower 157 nm optical absorbance also tend to show the longest lifetimes. The anti-proliferative activities of all the synthesized compounds were assessed on two different human cancer cell lines (Esophageal squamous cell carcinoma Ec9706 and Eca109), and the results showed that compound 4e showed the best anti-tumor activity with IC50 values of 8.23 mu M and 16.22 mu M against Ec9706 and Eca109 cell lines, respectively, which was 2-3 times more potent than 5-Fluorouracil (IC50 = 23.26 mu M and 30.25 mu M against Ec9706 and Eca109 respectively). Computations were carried out employing the restricted Hartree-Fock (RHF) and density functional theory (DFT) methods to investigate the geometries and energies for the 1,3-dithiole-2-thione (DTT), 1,3-dithiole-2-one (DTO), 1,3-dioxole-2-thione (DOT), and 1,3-dioxole-2-one (DOO) molecules and their radical cations. The geometrical parameters of all the four molecules suggest a finite extent of extended conjugation across the molecules. The radical cations of the DTT and DOT molecules exhibit increased extent of conjugation compared to their neutral analogues. However, on going from the neutral DOO and DTO molecules to their radical cations, the extent of conjugation across the ions decreases and two unsaturated sites become isolated from each other. Requirement of the relaxation energy for the formation of radical cation is much lower for the DTT and DOT molecules compared to that for the DOO and DTO molecules. Smaller relaxation energy requirement favors minimal resistance to charge transport along the molecular stacking. Therefore, it is speculated that the DTT and DOT molecules are better molecular conductors as compared to the DOO and DTO molecules.
